骨骼及牙齒為脊椎動物中兩個主要的礦化組織,而這兩個組織在發育分化過程中具有許多共通性,特別是在藉由細胞外基質(extracellular matrix, ECM)蛋白質來幫助整個組織礦化的過程。齒質母細胞(odontoblast)及骨細胞(osteoblast)在生長發育過程中皆會合成一些相同的細胞外基質(ECM),這些包括了兩大類的蛋白質:分別為膠原性蛋白(collagenous protein)及非膠原性蛋白(non-collagenous protein)。一般認為組織礦化的過程,乃是起於膠原蛋白形成網狀結構,而這些非膠原性細胞外基質蛋白會先磷化,繼而與鈣離子及磷酸結合,而形成結晶的核心,幫助羥基磷灰石(hydroxyapatite, Ca10(PO4)6(OH)2)晶體的形成及成長。因此,在脊椎動物骨骼及牙齒的發育過程中,礦物質的沉積及鈣化是最後一個重要的步驟。

牙本質基質蛋白-1(DMP-1;Dentin Matrix Protein-1)是參與牙齒及骨骼鈣化過程的一個非膠原性蛋白質。在骨生成 (osteogenesis)及齒質形成(dentinogenesis)過程,骨細胞(osteoblasts)與齒質母細胞(odontoblasts)會分泌非膠原性蛋白,並在細胞外形成基質(ECM),其中一類非膠原性的蛋白稱為SIBLING家族(SIBLING family;Small Integrin-Binding Ligand, N-linked Glycoprotein)。而牙本質基質蛋白-1(DMP-1)是這家族蛋白的一個成員。這些SIBLING家族的蛋白質具有許多共同的特性,例如,含有大量的唾液酸(sialic acid)、磷酸鹽成分,以及精氨酸-甘氨酸-天門冬氨酸(Arginine-Glycine-Aspartic acid;RGD sequence)細胞結合序列,此類酸性蛋白質在骨頭及牙齒組織的形成及礦化時會被分泌到細胞外基質(ECM)與羥基磷灰石(hydroxyapatite)結合,幫助羥基磷灰石晶體形成與組織礦化。

本研究的目標,以綠色螢光蛋白GFP為報導基因,以基因轉殖方法在斑馬魚及小鼠模式動物中分析人類DMP-1啟動子活性。結果顯示:在斑馬魚的骨細胞內,人類DMP-1啟動子的上游2,783 kb片段是有作用的,因為在過渡性的轉殖實驗結果中,發現Hu-DMP-1-GFP-2.783K-4及Hu-DMP-1-GFP-2.783K-36這兩個結構體可驅使報導基因綠色螢光蛋白在幼魚孵化後一個月的魚體上看到:頭部骨骼、鰓蓋、胸鰭、腹鰭、鰭骨、脊柱、鱗片這些部位有表現。同時,Hu-DMP-1-GFP-2.783K-36結構體,也可驅使報導基因綠色螢光蛋白於胚胎15.5天(dpc)及出生後在小鼠(F1)的骨細胞、牙齒及毛囊表現。由此結果顯示,在人類、小鼠、斑馬魚之間,此基因的調控機制具有保守性。
