旋光性的研究,在化學、生物領域,持續不斷的一直有很多科學家投入研究,由固態到液態並更進一步到氣態的旋光性量測。我們實驗室架設一套旋光角度量測系統能精確的量測旋轉角度,希望能有效的應用在基礎醫學或臨床醫院上面,由於人體組織對光光而言具有高度的散射性質,使得量測人體旋光性的靈敏度難以突破生理濃度而無法有效的進行臨床應用。
本論文提出以Zeeman穩頻雷射為系統光源之共路徑光學外差干涉儀,藉由這個光學架構,我們可以在散射介質其散射顆粒大小為1.072 、散射介質體積濃度為4.6%的溶液中成功的量出不同濃度的旋轉角度變化。本方法主要提出極化光子對『polarized photon pairs(PPPs)』的概念產生外差干涉的訊號,同時,利用實驗架構具polarization gating 和 spatial coherence的功能,而能有效的降低散射效應,提高旋轉角度量測的靈敏度,本實驗方法已成功的達到在4.6%體積濃度的散射溶液中測得11mM濃度的葡萄糖溶液達到生理濃度的要求。
A novel amplitude sensitive balanced detection polarimeter is setup in which a Zeeman He-Ne Laser and a differential amplifier are adopted in order to measure the optical rotation of glucose solution in a scattering medium. The minimum detectable concentration of glucose at 11mM in a 4.6% vol. concentration of polystyrene microsphere (1.072 ) scattering medium is measured successfully. In this polarimeter, the polarized photon pairs (PPP) from Zeeman laser presents a feature of common-path propagation in the scattering medium where the scattered PPP contributes the heterodyne signal coherently. In the mean time, the polarization gating and the spatial coherence gating of the polarimeter reduce the scattering effect also. In addition, the reduction of excess noise of laser intensity fluctuation results in a higher detection sensitivity of optical rotation measurement. The detection sensitivity in a nonscattering chiral medium is calculated and discussed.
