細胞膜是一個維持細胞生理一個很重要的關鍵,很多的分子在細胞膜表面有互動,全反射式螢光顯微鏡提供了一個良好的實驗平台來偵測細胞膜附近的動態行為,因為全反射式螢光顯微鏡的光學特性,可以大大的提高影像的訊號對雜訊的比值,在本研究中,我們利用全反射式螢光顯微鏡來偵測細胞膜附近的細胞骨架以及胞吞作用的液泡之間的交互作用,我們的結果顯示,利用全反射式螢光顯微鏡可以成功的來觀察細胞膜附近的動態影像,包括了微管(microtubule)的動態影像(dynamic instability),細胞骨架(actin,microtubule)及胞吞作用的液泡之間的交互作用。
Molecular interactions taken place near or on the cellular plasma membrane play an important role in maintaining cellular physiology. Moreover, molecular interactions on the plasma membrane are often expected to have different properties from those occurring in bulk solutions. Fortunately, total internal reflection fluorescence microscopy provides a good way to probe these important biological processes near plasma membrane with a much higher image quality than other optical microscopy. Using TIRFM, we can visualize the interactions between the cytoskeleton and the endocytic vesicles near the cellular growth substrate simultaneously at a higher signal to noise ratio.
