熱中風會引發腦缺血和神經細胞的死亡。本實驗利用光學顯微鏡、電子顯微鏡、免疫細胞化學染色法與西方墨漬法來探討大白鼠熱中風誘發後齒狀回分子層、顆粒層、多形層與海馬回CA4神經細胞與星狀膠細胞之形態變化和BDNF的相互關聯性。將大白鼠麻醉後置入恆溫箱中,應用生理多功能紀錄器連續監測大白鼠平均動脈壓與肛溫,當大白鼠處於42℃高溫環境時,平均動脈壓由最高點瞬間下降25㎜Hg 而且下降是不可回復的,此時界定為熱中風生成點。在光學顯微鏡下觀察,發現熱中風起始後0分鐘齒狀回顆粒層與海馬回CA4的神經細胞有受損的情形,但隨著時間的增加受損的情形趨漸緩和。以穿透式電子顯微鏡觀察,熱中風起始後0分鐘齒狀回顆粒層出現梭狀電子緻密度高的顆粒細胞而且有較長的細胞核而核內有空泡與明顯的核仁。而在顆粒層下區,電子緻密度呈現三角形與扁長形的神經細胞有明顯膨大的內質網與胞膜內空泡。在齒狀回分子層、多形層與海馬回CA4中,熱中風起始後5分鐘,GFAP-like-IR和BDNF-like-IR明顯地減少。隨著時間的增加,GFAP-like-IR和BDNF-like-IR也逐漸地增加。在齒狀回顆粒層GFAP-like-IR和BDNF-like-IR沒有明顯的變化。西方墨漬法分析海馬體BDNF蛋白質表現在熱中風起始後5分鐘劇烈地減少。因此,在熱中風後齒狀回和海馬回CA4中BDNF與星狀膠細胞可能參與神經細胞的回復與保護的機制。

Heatstroke induces cerebral ischemia and neuronal damage. BDNF-like immunoreactivity and the morphological changes were investigated by employing light microscopy, electron microscopy, immunocytochemistry and Western blotting in neurons and astrocytes of the dentate gyrus and hippocampal CA4 after heatstroke-induced rats. Heatstroke was induced by a high ambient temperature, until the mean arterial pressure began to decrease from the peak level. Pulsatile arterial pressure and colonic temperature were monitored continuously with a pressure transducer and a chart recorder (Polygraph Gould Modle 2107) during inducing heatstroke. In the granular layer and hippocampal CA4, neuron damage was found at the onset of heatstroke, and then decreased following different time courses. Under the electron microscope, the electron-dense granule cells had spindle-shaped outline and elongated nucleus containing intranuclear vacuoles and a conspicuous nucleolus in the dentate gyrus at the onset of heatstroke. In the subgranular zone, electron-dense triangular or elongated neurons had markedly dilated cisternae of the endoplasmic reticulum and marked hypertrophy of subplasmalemmal vacuoles. In the polymorphic and molecular layers of the dentate gyrus and hippocampal CA4, GFAP-like-IR and BDNF-like-IR markedly decreased at 5 minutes after heatstroke, and then gradually increased at 30 minutes after heatstroke. In the granular layer of the dentate gyrus, GFAP-like-IR and BDNF-like-IR remained unchanged through the different time courses of heatstroke. Using Western blotting, BDNF protein markedly decreased at 5 minutes after heatstroke. The data suggests that the BDNF and the astrocyte may be involved in the recovery and protection of the neuronal cells.
