果蠅的複眼由750-800個小眼所組成,在早期發育時期已知有一基因eye gone (eyg) 為複眼發育所必需。eyg突變會造成複眼的縮小甚至消失。相反的,將eyg做異位表現時,可以看到有額外的小眼生成。在果蠅基因體序列中,發現有一個與eyg 蛋白質序列相似的基因 (命名為twin of eyg,簡稱toe)。toe的表現區域與eyg相似,異位表現也可以造成額外小眼的生成。為了更進一步探討toe的功能與這兩個基因之間的關係,需要有單一基因突變或雙重突變的基因背景。因此,在我的論文中,我試著利用基因剔除的方法來製造toe完全突變(null mutant)的果蠅。另外,我利用跳躍子誘發的染色體互換來試著製造並篩選出eyg與toe的雙重突變。另一方面,我的論文研究了Notch訊號對於eyg和toe的調控機制。發現eyg和toe在眼盤赤道線和前方背側的表現,需要Notch來活化。而這作用乃是藉由eyg-toe基因座中兩段分別命名為B8和E2XS的調控子來達成。
During the early development of Drosophila compound eye, several genes are known to be required, and eye gone (eyg) is one of them. Loss-of-function mutations of eyg lead to reduction or absence of adult eye. Ectopic expression, however, can induce ectopic eye in the ventral side. The loss-of-function and gain-of-function phenotypes suggest that eyg acts as an important regulator of eye development. A predicted gene with high sequence homology to eyg was identified from the fly genome sequence. We named it twin of eyg (toe). The two genes have nearly identical expression pattern and gain-of-function phenotypes. To analyze the function of toe and the relationship between eyg and toe, null mutants of eyg and toe respectively are needed. Having eyg mutants already, in this thesis, I used the targeting method to hunt for the toe null mutant. Besides, I also tried to generate the eyg and toe double mutant by male P-element recombination.Furthermore, I analyzed the molecular mechanism of eyg induction by Notch signaling. In larval stage, eyg/toe can express in the equatorial region and anterior dorsal part of eye disc. In this thesis, I’ve shown the expression required the activation by Notch signaling through two cis-elements of the eyg-toe locus, B8 and E2XS.
