沾黏(Adhesion)是外科手術時常會遇到的問題,抗沾黏膜是最常用的預防方法。本實驗室過去曾以聚半乳醣醛酸(PGA)來製備抗沾黏膜,有很好的抗沾黏效果;而本研究希望更進一步改善PGA抗沾黏膜,添加蘆薈多糖(acemannan)抗發炎活性成分,和PGA混合並以carbodiimide來交聯製備抗沾黏膜。當蘆薈多醣所佔之比例愈高,抗沾黏膜的含膠量下降,含水量上升,而交聯之間數目平均分子量(Mc)上升。可得知蘆薈多醣所含之比例愈高,膜交聯的效果愈差。由動物實驗的結果可知,PGA和P7A3 (PGA70%/acemannan30%)膜其抗組織沾黏的情形較市售抗沾黏膜Seprafilm好。經由腹水細胞數測量的結果,可以得知添加蘆薈多醣的P7A3膜比PGA膜,於動物實驗結果上引起的發炎現象較輕微。切片觀察的結果可以知道Seprafilm分解較快,七天已完全看不到膜存在;而PGA、P7A3這兩組,其分解的速度比較慢,七天時可觀察到材料的存在,而十四天時則完全看不到膜的存在。
由本研究的結果顯示,添加蘆薈多醣的P7A3膜,和PGA膜都具有良好的抗沾黏效果,而P7A3膜於動物實驗結果上引起的發炎現象較PGA膜輕微,藉此可以有效地改善PGA抗沾黏膜導致發炎的現象。
