隱胞子蟲症是一種由隱胞子蟲(Cryptosporidium spp.)寄生於各種動物所引起的原蟲性疾病,也是一種人畜共通傳染病。本類寄生蟲可引發人畜之下痢症狀,在免疫機能不全的人畜甚至可造成死亡。本研究自九十年九月至九十二年十月間採集國內19個馬場共計436個馬糞便檢體,以抗酸性染色法進行隱胞子蟲的調查。總計檢出161個檢體是隱胞子蟲陽性,陽性率為36.9%。自陽性檢體中選取33個進行巢式聚合酶連鎖反應,限制酶切割圖譜及核酸定序等分子生物學技術分析。結果其中31個檢體為牛型之C. parvum;另外2個從北部地區及甫自國外輸入的馬匹檢體中發現了增幅之核酸片段(605bp)與C. felis吻合的隱胞子蟲,但因未對其進行病理、交叉感染、病原性及致病力…等各種生物特微之研究,因此尚無法證實。而牛型的C. parvum及C. felis均屬人畜共通,值得注意。本研究以抗酸性染色法檢出陽性檢體,再以GIT方法抽取檢體中隱胞子蟲的DNA進行巢式聚合酶連鎖反應,產物以限制酶分析區分種別,進而以核酸定序鑑別出馬隱胞子蟲的基因型態,此模式可作為日後運用分子生物學來調查或診斷馬隱胞子蟲基因型別之參考。
Cryptosporidiosis is a zoonotic disease caused by the protozoan parasite Cryptosporidium spp. This parasite can cause diarrhea in animals and humans, even cause death in immunosuppressed patients. The survey was conducted during Sept. 2001 to Oct. 2003 to study equine cryptosporidiosis. A total of 436 fecal samples were collected from 19 farms and method of acid-fast stain was used for primary screening. Oocysts of Cryptosporidium were recovered from 161 samples and the prevalence rate was 36.9%. Among positive samples, 33 were selected for further analysis by nested PCR, RFLP, and nucleotide sequencing. The result showed that 31 of them were C. parvum, bovine genotype and the other two, one each from farms of northern Taiwan and imported, having amplified nucleotide fragment(605bp) which to be identical to C. felis. However, further confirmation is required to assure the infection of C. felis in equine. Nevertheless, the fact that C. parvum, bovine genotype and C. felis are zoonotic makes it worth paying attention to. In this study, a system containing a primary screening test by acid-fast stain, followed by extraction of DNA with GIT method, amplification of DNA by nested PCR, analyzation by RFLP, and finally used identification by nucleotide sequencing may be used as a tool for analysis of genotype in equine cryptosporidiosis.
