論文名稱:台灣地區漢人粒線體DNAHV1與HV2區異質型之分析

中文摘要
粒線體DNA具母系遺傳、序列鹼基多型性及每個細胞內存在多套基因組等特性,使得核DNA即使裂解嚴重或DNA含量過少時,仍可進行粒線體DNA之分析比對,然而同一個體不同組織之樣品其粒線體DNA有時會出現異質型(heteroplasmy),也因此影響著粒線體DNA鑑定結果的詮釋,故本研究乃分析53個個體各5種部位之樣品(含唾液、血液、毛髮、尿液及指甲)其粒線體D-loop之HV1及HV2區之序列,共發現3個個體樣品存在異質型,出現異質型位點包括HV1區的L16179位點(T/C)、L16093位點(T/C)及HV2區的L195位點(T/C);而出現異質型個體之毛髮及指甲經重新收集樣品、萃取DNA及序列分析後,則發現原發生於毛髮HV1區之L16093的T/C異質型序列於另一根毛髮卻出現C的同質型,而另外一個發生於毛髮HV2區之L195的T/C異質型序列於另一根毛髮亦出現T的同質型,且另外一個發生於指甲HV2區之L195的T/C異質型經重新採樣分析亦出現T的同質型;因此,建議在刑事鑑識上倘若證物或已知來源者之樣品出現異質型,可再收集其他組織或同一組織其他樣品,以解決鑑定結果詮釋上的困難。本實驗針對高解析熔解(High-resolution Melting)多型性分析應用在粒線體DNA鑑定上做評估。熔解曲線不同時可確定其序列一定不同,當熔解曲線相似時則無法確定其序列是否存在差異,因此,針對這些樣品應再進行序列分析加以確認,但利用此技術仍可達到粒線體DNA母系遺傳初步篩檢的目的。
