本論文有鑑於近年來許多研究指出,富含反式脂肪酸的部分氫化油被認為可能會提高血漿中膽固醇濃度,然而也有其他的研究提出不同的結果。為了釐清反式脂肪酸對血脂質的影響,本研究以交替模式長期飼養來探討反式脂肪酸對於血脂濃度之影響。以雄性SD (Spague Dawley)大白鼠為實驗動物,共分兩組,實驗時間為19週,實驗期間採自由攝食、飲水。將實驗週期分為三階段,分別是:第一週期A組攝食含添加10%部分氫化油(富含反式脂肪酸),B組攝食含添加10%橄欖油(富含順式脂肪酸),為期六週。經一週之調整飲食(Washout)後進行第二週期實驗,A組攝食含添加10%橄欖油(富含順式脂肪酸)、B組攝食含添加10%部分氫化油(富含反式脂肪酸)為期六週。再經一週之調整飲食(Washout),進行第三週期無膽固醇攝食模式,飼料中移除膽固醇與膽鹽,A組攝食含添加10%部分氫化油(富含反式脂肪酸),B組攝食含添加10%橄欖油(富含順式脂肪酸),為期五週。實驗期間每週測量一次體重,每兩週採取一次尾靜脈血以分析其血漿脂肪膽固醇以及三酸甘油脂與脂蛋白組成份。實驗結果發現,第一週期血漿總膽固醇分析顯示:攝取部分氫化油組之血漿總膽固醇濃度明顯低於攝取橄欖油的組別,而此現象可能與極低密度脂蛋白膽固醇濃度變化有關。而此趨勢經交替飲食後亦會出現在第二週期。第三週期兩組間之血漿脂質與之脂蛋白值皆無統計上顯著差異。此外,在washout期間,兩組血脂質卻都沒有統計上的差異。綜合以上結果推測,反式脂肪酸對血膽固醇濃度的影響與否可能與飼料中是否有膽固醇的添加有關。
Abstract
Lots of researches point out, some hydrogenated oil containing trans-fatty acid can be thought to increase the concentration of cholesterol in blood, yet other studies proposed different results. To further clarify the impacts of trans-fatty acid on the fat in the blood, this research is to probe into the influences of trans-fatty acid on the concentration of fat in the blood by raising rats with the cross-over method in a long period.

The male SD (Spague Dawley) rat were used as observant and divided into two groups. The period of experiment lasted for 19 weeks. Fresh diets and water were available ad libitum. The experiment was conducted by three stages. Initially, in the first period of six weeks, group A was bred with stuff made of 10% of the hydrogenated oil (rich trans-fatty acid included), and group B with forage comprising 10% of the olive oil (substantial cis form fatty acid included). After the first phase, one week was undertaken in which the diet adjustment (Washout) was carried out. Next, group A was fed with materials in which 10% of the olive oil (substantial cis form fatty acid included) was added, and group B with substance including 10% of the hydrogenated oil (rich trans-fatty acid included) during the second period of six weeks, followed by another one week of diet adjustment (Washout). Finally, in the third period of five weeks, the diets were similar to those in the first step but with no cholesterol and cholic acid added.

The results of this study indicated that, in the first period, the concentration of cholesterol of group A, taking up the diet rich hydrogenated oil, was higher than group B that had diet rich olive oil. This could be caused by the variation of concentration of VLDL. For the second step, the concentration of cholesterol of group B assimilating the diet rich hydrogenated oil was higher than group A absorbing diet rich olive oil. In the third phase, the concentration of cholesterol showed no significant difference. According to the findings above, a supposition is proposed that, the relationship between trans fatty acids and the level of cholesterol in blood may be correlated with the cholesterol whether added or not.
