研究指出茄紅素(Lycopene)具有良好的抗氧化功效。本研究目的為探討茄紅素的給予對老化促進小鼠學習記憶能力與體內抗氧化狀態之影響。實驗以三月及六月齡老化促進小鼠(senescence accelerated mice, SAMP8)為對象,分為對照組、4、20及100 ppm茄紅素組,為期12週。實驗期間記錄攝食量、攝水量、體重變化及老化指數之評估。在行為測試方面進行活動量試驗,以主動迴避試驗及單次被動迴避試驗評估學習記憶能力。犧牲後,進行血液生化分析,檢測肝臟中超氧歧化(SOD)、觸(catalase)活性以及丙二醛(MDA)與總含硫化合物含量。以組織切片觀察腦部β-類澱粉蛋白沉積之情形,另外觀察精子濃度及活動力。結果顯示實驗各組攝食量、體重變化及活動量於四組間均無顯著差異;而攝水量方面除了三及六月齡雌性SAMP8小鼠外,茄紅素組與對照組相比為顯著增加(p<0.05)。在老化指數評估方面,六月齡雄性與雌性小鼠飲用100 ppm茄紅素組之老化指數總分顯著低於對照組(p<0.05)。於學習記憶方面,經主動迴避試驗結果發現三及六月齡雄、雌性小鼠實驗組均有較佳的學習記憶能力;另三及六月齡雄、雌性小鼠經單次被動迴避試驗發現100 ppm茄紅素組其在學習記憶保留上顯著較佳,且達顯著差異(p<0.05)。肝臟SOD方面,六月齡雄性小鼠20、100 ppm茄紅素組顯著高於對照組(p<0.05);六月齡雌性小鼠100 ppm茄紅素組顯著高於對照組(p<0.05)。三月齡雄性小鼠100 ppm茄紅素組之肝臟catalase活性顯著高於對照組(p<0.05);而6月齡雄性小鼠20 ppm茄紅素組及雌性茄紅素實驗各組之catalase活性與對照組比較皆顯著增加(p<0.05)。六月齡雄性小鼠20、100 ppm茄紅素組之肝臟MDA生成量比對照組顯著降低(p<0.05)。於腦部β-類澱粉蛋白總沉積量方面,三月齡雌性小鼠20、100 ppm茄紅素組比對照組顯著降低(p<0.05);六月齡雄性小鼠4、100 ppm茄紅素組及雌性20、100 ppm 茄紅素組之β-類澱粉蛋白總沉積量比同性別對照組顯著降低(p<0.05)。此外,在精子活動力及濃度方面,實驗各組間均無顯著差異。綜合以上結果,補充茄紅素可有效改善SAMP8系小鼠學習記憶能力減少腦部病變,藉由降低氧化傷害而延緩老化發生。
Lycopene was demonstrated to has antioxidant capacity. The purpose of this study was to investigate the effect of lycopene on learning memory ability and antioxidative status in senescence accelerated mice (SAMP8). Three and six month-old SAMP8 mice were divided into four groups: control group、4、20、100 ppm lycopene(experimental group). After 12 weeks of feeding, body weight, food intake, drink amount, aging score, open field activity test, active shuttle avoidance test and single-trial passive avoidance were performed during the experiment. The biochemical parameters of serum were analyzed after sacrificed. The activities of superoxide dismutase (SOD), catalase and malondialdehyde (MDA), total thiol concentrations in the liver and pathological examination of brain, motility and concentrations of spermatozoa in male mice were also evaluated. The results showed that there were no significant differences in the body weight, food intake and locomotion among four groups in both 3 and 6-month old mice. The drink amount of experimental groups were higher than the control group in both 3 and 6-month old male mice (p<0.05). The aging score of experimental groups were significantly lower than the control group in the 6-month old mice (p<0.05). For the 3 and 6-month old mice, experimental groups had better active shuttle avoidance response;3 and 6-month old mice drink with 100 ppm lycopene had significantly better single-trial passive avoidance results (p<0.05). In 6-month old male mice, the SOD activity of liver, 20、100 ppm lycopene groups were significantly higher than control group. In 6-month old female mice, the liver SOD activity, in the 100 ppm lycopene group was significantly higher than that of control group (p<0.05). In 3-month old male mice, the liver catalase activity, in the 100 ppm lycopene group was significantly higher than that in the control group (p<0.05). The liver catalase activity of 20 ppm lycopene group in the 6-month old male mice and the experimental groups in the 6-month old female mice were significantly higher than that in the control group (p<0.05). The MDA concentrations of liver in the 20、100 ppm lycopene groups in the 6-month old male mice and 4、100ppm lycopene groups in the 6-month old female mice were significantly lower than control group. The β-amyloid protein deposition of brain in the 20、100 ppm groups were significantly lower than control groups in both 3 and 6-month old female mice;in the 4、100 ppm lycopene groups were significantly lower than control groups in the 6-month old male mice. The motility and concentrations of spermatozoa were no difference in both 3 and 6-month old male mice. From the findings of these results, the supply of lycopene may attenuate the age-related learning impairment and reduce pathological changes of the brain in SAMP8 mice. It’s suggested that lycopene supplementation may promote the antioxidative ability to lowering the oxidative injury.
