本研究探討攝取甘薯葉對於膽固醇飲食之倉鼠(Syrian hamsters)血脂質、體內自由基及造成血栓時間之影響。將甘薯葉(Sweet potato leaf)磨成甘薯葉粉 (Sweet potato leaf powder,以SPLP表示),添加於含膽固醇之飼料中,實驗分為六組,分別為攝取基礎飼料之C組及於基礎飼料中分別添加0.1%膽固醇之E組; 0.2%膽固醇之F組; 0.1%膽固醇+2.5%SPLP之A組; 0.1% 膽固醇+5%SPLP之B組;0.2%膽固醇+5%SPLP之D組。均採自由給食,實驗為期六週。結果顯示,A、B組血漿總膽固醇、VLDL-C均顯著比E組低,D組之總膽固醇、LDL-C均顯著的比F組低。在促動脈硬化指標(Atherogenic Index;AI)值方面顯示D組顯著較F組約低37.6%。在總三酸甘油脂(Total Triglycerol;TG)顯示,A、B兩組顯著均較E組低,D組顯著較F組低。在中性糞便固醇含量的變化方面,餵食甘薯葉之各組皆顯著比其他組高。在離體自由基方面顯示,餵食甘薯葉之各組皆顯著較低,約降低66~93%。利用15%氯化鐵傷害模式以誘導血栓的形成,結果發現B、D兩組所需的時間較長,B組比A組長4.11倍;D組比F組長3.6倍。對於各組倉鼠體內抗氧化酵素含量以Western blot分析,顯示餵食甘薯葉對於倉鼠體內的抗氧化酵素及LDL-R表現量均有較多的現象。綜合以上結果得知,甘薯葉因可增加糞便中性固醇排出量及LDL-R的活性,故具有調降血漿總膽固醇及LDL-C的含量之現象,又甘薯葉因具有活化肝臟內抗氧化酵素Superoxide Dismutase(SOD)等之活性,降低體內自由基含量並延緩氧化鐵誘導血栓傷害所需之時間,具有保護心血管之作用。
This study was designed to explore the effect of sweet potato leaf powder (SPLP) feeding on the parameters of plasma lipids, free radicals and time to thrombsis formation in Syrian hamsters fed on hypercholesterolemic diets. The animals were separated into six groups: feeding control diet (group C), control diet supplemented with 0.1% cholesterol (group E), with 0.2% cholesterol (group F), 0.1% cholesterol + 2.5% SPLP (group A), 0.1% cholesterol + 5% SPLP (group B), and 0.2% cholesterol + 5% SPLP (group D) for six weeks. The results indicated that the level of total cholesterol and very low-density lipoprotein cholesterol (VLDL-C) was significantly reduced in groups A and B than in group E. The level of total cholesterol and VLDL-C in group D was also lower than that in group F. Atherogenic index (AI) in group D was singnificantly decreased (37.6%) than that in group F. The triglyceride (TG) level in groups A and B singnificantly lower when compared to group E, whereas the TG level was also reduced in group D when compared to group F. SPLP feeding increased fecal sterol contents. SPLP decreased free radicals production (66-93%). SPLP feeding elongated ferric chloride induced time to thrombosis formation (3.6-4.1 folds). SPLP feeding increased liver superoxide dismutase contents and low-density lipoprotein receptor (LDL-R) expression. In summary, SPLP feeding can reduce plasma lipids by the increased LDL-R expression and fecal sterol contents. SPLP feeding can upregulate hepatic superoxide dismutase contents and consequently delay 15% ferric chloride-induced arterial thrombosis formation. These results suggest that SPLP feeding maybe used to prevent cardiovascular diseases.
