檸檬香茅(Cymbopogon citratus),就是廣為人知的檸檬草,在台灣被人們栽種並作為民間草藥,也是養生茶中常見的一味草藥。本篇研究重點在於研究檸檬香茅對離體豬冠狀動脈的血管舒張作用,並探尋其有效成分究竟為何。首先,製備檸檬香茅之甲醇層粗萃取物(ME),以此萃取物進行分離所得的五個有機溶媒層,如水層萃取物(WtF)、正丁醇層萃取物(BtF)、乙酸乙酯層萃取物(EaF)二氯甲烷層萃取物(DcF)和正己烷層萃取物(HxF)。然後將這些萃取物來進行血管舒張作用的評估。經過實驗之後發現除了WtF 之外,ME、HxF、DcF、BtF和EaF 在濃度1 mg/ml時都具有明顯的冠狀動脈舒張作用。在這些活性有機溶媒層中又以HxF活性最強,IC50=0.084±0.027(mg/ml)。因此我們更進一步以HxF來探討各種機轉,如去除血管內皮、抑制NO合成、對鈣離子內流的影響、抑制KATP通道及β接受體測試。結果顯示,HxF可能是經由內皮依賴(Endothelium-dependent)、促進內皮NO的合成並活化cGMP-independent pathway及抑制鈣離子湧入等機制來達到舒張冠狀動脈的作用。根據文獻記載,Citral與Geraniol是檸檬香茅中的主要成分之ㄧ,也具有多種生物活性。在氣相層析儀的分析結果中,更證實了此二化合物的存在。經冠狀動脈舒張活性試驗後也發現它們確實具有血管舒張活性。在舒張機轉方面,除了Geraniol對高鉀所引發的鈣離子內流沒有抑制作用之外,還發現Citral可以降低細胞內鈣離子濃度,Citral與Geraniol之舒張機轉大致與HxF相同。這更證明Citral與Geraniol確實是HxF中的活性成分之一。
Cymbopogon citratus as worldly known as lemongrass has been cultivated in Taiwan as folk medicine and component in herbal drink for health purpose in Taiwan. The present study investigates the vasorelaxation effect of lemongrass in isolated porcine coronary artery. The methanolic extract (ME) and its five fractions of water (WtF), butanol (BtF), ethyl acetate (EaF), dichloromethane (DcF), and n-hexane (HxF) were prepared and then subjected to vasorelaxation evaluation. The results revealed that ME, HxF, DcF, BtF and EaF possessed significant relaxation effect in porcine coronary artery at the concentration of 1mg/ml with the exception of WtF. Among these active fractions, HxF, EaF and DcF showed fully vasorelaxation activity. Especially the HxF, IC50 was 0.084±0.027 (mg/ml)achieved. So we performed mechanism studies by HxF. For example, endothelium denudation, inhibition of NO synthesis, influence to calcium influx, inhibition of KATP channel, and β-receptor. The result indicated that HxF relax coronary artery by endothelium-dependent pathway, promotion of NO synthesis, activation of cGMP-independent pathway and inhibition of calcium influx. According to the literature, citral and geraniol were two of the major component of lemongrass and possessed variety bioactivity. We proved the presence of citral and geraniol by gas chromatography. Besides geraniol without activity on calcium influx, citral can reduce [Ca2+]i. They can relax the coronary artery by the same machenisms. The result revealed that citral and geraniol were two of the active components in HxF.
