哺乳動物的毛囊是一種動態結構,經由生長、重朔和掉髪的週期調控過程形成髮幹。毛髮週期歷經三個階段:生長期(anagen),消褪期(telogen)及休止期(catagen)。這些轉變是藉由環境中訊息的改變所調控,如細胞激素、賀爾蒙、神經傳導物質的變化和活化以及其毛囊生長相關酵素接受器、轉錄因子和與其受器的變化酵素的作用。基質金屬蛋白酶(MMPs)是ㄧ個需要鋅離子的中性蛋白酶家族,為解離細胞外基質和基底膜蛋白質的主要酵素,可使細胞進行移行、分化及發炎等反應。在我們最近的實驗中發現MMP27的訊息核糖核苷酸位在毛囊的外根鞘中。本論文的目的即是要探討MMP27的表現與毛髮週期兩者之間的關係。在脫毛動物模式中利用免疫組織化學染色後發現MMP27蛋白質持續表現在消退期至生長期,並且由西方墨點法也得知MMP27蛋白質在消退期中的表現量最高,但在生長第六期(anagen 6)的表現最低。此外,在脫毛後小鼠的背上塗抹MMP的抑制劑(GM1489)後發現,從生長第二期(anagen 2)到生長第六期(anagen 6)其部分毛囊的生長有明顯抑制效果。這些結果顯示,在毛囊生長的過程中MMP27可能參與了調控的機制。
The mammalian hair follicle is a dynamic structure that generates a hair shaft through a tightly controlled cycle of growth, remodeling, and loss. Hair cycling is the rhythmic change of the hair follicle (HF) through three phases: anagen (follicle generation and hair production), catagen (follicle regression), and telogen (resting phase). These transformations are controlled by changes in the local signaling milieu, based on changes in expression/activity of a constantly growing number of cytokines, hormones, neurotransmitters, and their cognate receptors as well as of transcription factors and enzymes that have become recognized as key mediators of hair follicle cycling. Matrix metalloproteinases (MMPs), a large family of zinc-dependent, neutral proteinases, are major components of the enzyme cascade responsible for degradation of extracellular matrix and basement membrane proteins that participate in cell migration, differentiation and inflammation. In our recent founding MMP27 mRNA was detected in hair’s outer root sheath (ORS) . The purpose of this study was to demonstrate the correlation between hair cycle and the expression of MMP27. The MMP27 protein was expressed from telogen to anagen in our depilation mouse model by immunohistochemistry. Temporal expression of MMP27 during hair growth was further demonstrated by western bloting. Moreover, treatment with MMPs inhibitor (GM1489) in mice skin after depilation revealed a significant inhibition of HF growth from anagen 2 to anagen 6. These results suggest the potential role of MMP27 in participating HF growth.
