濱當歸(Angelica hirsutiflora)為當歸屬多年生的大型草本植物。將濱當歸甲醇粗抽物經藥效篩選後,於抗糖尿病藥理試驗中,發現濱當歸粗抽物在細胞與動物試驗中,均具有促進胰臟蘭氏小島β-細胞生成更多胰島素與降低第二型糖尿病小鼠血糖濃度與增加胰島素敏感度的效果。抗氧化方面,發現濱當歸之正丁醇可溶層有清除自由基的作用。另外,於抗發炎藥理測試得知濱當歸甲醇粗抽物、氯仿可溶層與正丁醇可溶層的IC50值均低於10 μg/ml,其對以fMLP/CB刺激嗜中性白血球所引發的超氧自由基釋放與elastase釋放有抑制作用,因此對濱當歸之化學成分與生物活性作詳細的研究。
將濱當歸粗抽物以氯仿、正丁醇和水進行多次分配萃取後,共分得三層。將各分配層進行管柱層析後,共純化得到41個化合物。其中以香豆素類與色酮類化合物為主要成分。再將所分離得的純化合物進行藥理試驗後,發現化合物rutin與protocatechuic acid具有清除DPPH自由基的抗氧化作用。於抗發炎的測試中,則發現到化合物osthol、auraptenol、bergapten、imperatorin與 (2”S)-hamaudol能抑制超氧自由基釋放與elastase釋放。而抗糖尿的活性測試尚在進行中。
Angelica hirsutiflora is a huge perennial herbaceous plant . The results of anti-diabetes experiments indicated that the crude extract of A. hirsutiflora significantly increased insulin secretion in a pancreatic islet β-cell and inhibited hyperglycemia situation, insulin sensitivity in high fat diet-induced diabetic mouse. The crude extract and its partitioned layers of A. hirsutiflora inhibited the release of superoxide anion and elastase in fMLP/CB-activated human neutrophils. The IC50 values of inhibition effects of superoxide anion and elastase release were lower than 10 μg/ml. The n-BuOH soluble layer also had the ability for reducing DPPH. Therefore, we are interested in the bioactive principles of A. hirsutiflora.
Forty-one compounds were isolated from the crude extract of A. hirsutiflora. Their structures of thirty-seven compounds were determined by spectral analysis. The major components are coumarins and chromones. Rutin and protocatechuic acid both showed significant effect for reducing DPPH. In the anti-inflammatory experiments, osthol, auraptenol, bergapten, imperatorin and (2”S)-hamaudol had the inhibition of inflammatory responses. The anti-diabetes assay of pure components are under proceeding.
