隨著生醫材料的研究與發展,幾丁質這個在天然有機物當中含量僅次於纖維素的多醣体,其重要性就更加受人矚目。而幾丁聚醣則是幾丁質去乙醯化後所得之產物,兩者皆具有良好的生物相容性及生物可降解性,臨床上不僅具有抗菌、止血、促進傷口癒合等特性,更是一個沒有毒性療效又好的牙周再生材料。四環黴素由於具抑菌功能,並能促進纖維母細胞的附著,以及抑制膠原蛋白的活性等功能,而且在牙齦溝液內較高濃度,因此常用於牙周病的治療。本研究以經由三聚磷酸交聯後之幾丁聚醣與四環黴素海綿體於體外釋放之期程及對細菌抑制之表現來探討臨床牙周疾病治療使用之可行性。四種經過不同處理之幾丁聚醣海綿體為本實驗之主要材料,包含:1. 單純幾丁聚醣海綿體為控制組;2. 經三聚磷酸交聯之幾丁聚醣海綿體(TPP);3. 未經三聚磷酸交聯之幾丁聚醣與四環黴素海綿體(TC);及4. 經三聚磷酸交聯之幾丁聚醣與四環黴素海綿體(TPP+TC)。海綿體之重量、厚度、半徑均予以測量,且均接受掃瞄電子顯微鏡(SEM)照相。以浸泡於蒸餾水中方式測量海綿中四環黴素於體外釋放之期程,同時分別將體外釋放四環黴素後之海綿體於細菌實驗室進行抑菌圈實驗。本研究結果顯示,經由三聚磷酸交聯後之幾丁聚醣與四環黴素海綿體可延長藥物釋放時間至11天,同時依然具有抑菌效果,而未經三聚磷酸交聯之幾丁聚醣與四環黴素海綿體雖然亦有延長藥物釋放時間與抑菌效果,但於7日內即崩解,其餘則均無藥物釋放時間與抑菌效果,故而經本研究之結果,經由三聚磷酸交聯後之幾丁聚醣與四環黴素海綿體可以進一步研究推廣至臨床使用。
