中文摘要

針對番荔枝科哥納香屬植物台灣產恆春哥納香(Goniothalamus amuyon)的莖部、根皮部、葉部及馬來西亞產Goniothalamus velutinus的莖部,進行化學活性成分分離。由G. amuyon莖部得到二十個styryllactones、十個alkaloids、四個steroids及六個acetogenins; 根皮部得到九個styryllactones; 葉部得到七個styryllactones。由G. velutinus莖部得到一個styryllactone、二個steroids、一個alkaloid及十個acetogenins。其中包括十個styryllactones: isogoniothalamin oxide (S3)、 8-methoxygoniodiol (S7)、 8-chlorogoniodiol (S9)、 digoniodiol (S10)、 deoxygoniopypyrone A (S12)、 goniothalesacetate (S14)、 goniofupyrone A (S15)、 methylgonioamuate (S16)、 methyldihydrogonioamuate (S17)及goniothalesdiol A (S18)為新化合物,此外有十三個化合物為首次自本屬植物分離得到。同時從中國大陸瓜馥木屬植物排骨靈(Fissistigma bracteolatum)得到包括十一個alkaloids、四個flavonoids及四個steroids類等十八個化合物,其中bractelactone (AF1)為新化合物。
在生物活性研究方面將styryllactones化合物進行抗腫瘤、抗血小板凝集及抗發炎測試,其中化合物goniothalamin (S1)及isogoniothalamin oxide (S3)對於HepG2、DU145、PC-3和HepJ5等癌細胞株具有顯著的細胞毒殺活性其IC50值分別為0.31、0.40、0.51、0.19及0.19、0.45、0.18、0.20 μg/ml,8-chlorogoniodiol (S9)對於HepG2 (IC50 0.64 μg/ml)癌細胞株具有顯著的細胞毒殺活性。化合物 goniothalamin (S1)和8-chlorogoniodiol (S9)的抗癌作用機轉正在進行研究中。此外將F. bracteolatum所得到包括bractelactone (AF1)、 piperlactam A (AF5)、 aristolactam BII (AF8)、 aristolactam BIII (AF9)、dehydrodiscretamine (AF11)、 isopedicin (AF12)、 5,8-dihydroxy-6,7-dimethoxyflavanone (AF13)及pinostrobin (AF14)等化合物進行抗發炎活性測試。其中isopedicin (AF12)對人類嗜中性白血球(neutrophils) superoxide generation具有抑制活性(IC50 0.34 μM)。
另外,除了建立具有抗癌活性的styrylpyrone類化合物 goniothalamin (S1)、goniothalamin oxide (S2)、isogoniothalamin oxide (S3)、goniodiol-7-monoacetate (S5)、goniodiol-8-monoacetate (S6)、 8-methoxygoniodiol (S7)及8-chlorogoniodiol (S9)指紋圖資料庫外,同時針對恆春哥納香各部位所含抗癌活性成分含量進行定性及定量分析。
