硫辛酸(alpha-lipoic acid,ALA)是存在於粒線體內的雙硫化合物,可由細胞自行少量產生。ALA 主要的生理作用為粒線體內 pyruvate dehydrogenase 之輔酶,亦為生物膜維持正常功能的組成要素。近年來的研究指出,ALA 是一種強力的抗氧化劑及自由基清除者;而 ALA 的還原態二氫硫辛酸(dihydrolipoic acid, DHLA)在研究中也被發現具有抗氧化能力。同時我們也知道氧化性壓力在許多發炎性疾病中扮演著很重要的角色。另有研究指出,ALA 可藉由增加 T 細胞內 cyclic AMP(cAMP)的濃度,以抑制 T 細胞的活化與遷移。在血小板的活化過程中,ROS 與 cAMP 皆扮演重要角色,所以我們推論 ALA 應有抑制血小板凝集的可能。然而,目前尚無此方面的報告。因此,本研究主要探討 ALA 對血小板凝集反應之影響。
本研究結果首度證實 ALA 確實能夠明顯抑制 arachidonic acid(AA, 100 µM)或 collagen(10 µg/ml)所引起的兔子血小板凝集反應且 IC50 分別為 400 µM 和 200 µM,其可能作用機轉包括(1)經由抑制 cyclooxygenase 及 thromboxane synthase 的活性而抑制 thromboxane A2(TxA2)的生成(2)透過刺激 adenyl cyclase 活性而增加血小板內 cAMP 的濃度(3)抑制細胞內鈣離子的釋放(4)增加血小板細胞膜的流動性(5)抑制血小板內ROS的生成等。
總結,本論文首度證明ALA具有抗血小板凝集之功能。希經由這些研究結果對未來ALA應用在抗凝血及心血管疾病等方面提供有價值的貢獻。
