本試驗探討國蘭雜交種‘18學士×春蘭’以液態培養的方式,添加不同種類的生長調節劑,對根莖生長分化之影響。在相同培養時間下根莖的芽體增殖數,液態培養遠大於固態培養。在液態培養中,不同的生長調節劑對根莖分化有明顯的差異,細胞分裂素中的TDZ與BA可使根莖誘導為芽體,而植物生長素NAA則可促進根莖增殖。利用TDZ、BA等生長調節劑進行根莖之芽體增殖試驗,可發現使用0.01㎎/L TDZ的液態培養,其芽體數為多,芽體成叢生狀,但芽體形狀較小;使用3㎎/L BA 2個月後,其芽體數較少,但芽體較大。根莖之芽體增殖率以TDZ相對於BA為高,就芽體生長速度而言,以BA有較高的助益。以BA與TDZ誘導液態培養所得之根莖,其芽體數與芽體長皆優於固態培養。在根莖增殖根莖的實驗中,則發現NAA對根莖的增殖和分枝的增加有顯著的效果。誘導出的芽體有其發根困難,生長緩慢的問題。BA與NAA進行發根實驗,發根情形不良。以蔗糖與MS進行液態培養,芽體發根率優於固態培養,且無根莖的形成。
