摘要
本研究藉由粒線體DNA(mtDNA)多型性的變異,來探討廣泛分佈於台灣海拔兩千八百公尺以上的台灣冷杉(Abies Kawakamii)之親緣地理關係。本研究選取大霸尖山、雪山主東峰、北二段無名山、合歡東峰、奇萊南華、玉山、關山等八個樣區,選取67個單株進行研究,研究中共測試十八對Primer,只有兩對Primer FO303/FO304及nad5/1-2a可以呈現單一Band,序列比對後發現這兩段基因序列毫無變化。除了冷杉推測可能與台灣冷杉正處於冰河時期過後往高海拔退縮,族群正面臨瓶頸現象有關。
The spatial pattern of mitochondrial DNA(mtDNA)variation in Abies kawakamii (Hayata) Ito(Pinaceae)was studied in 67 trees collected in Taiwan Alpine(8 population).By sequencing 2 mtDNA intron fragmens using 2 primers(FO303/FO304 and nad5/1-2a).We revealed a total of 2675bp with no polymorphic site(FO303/FO304:746bp , nad5/1-2a:1929bp).Among them, there is no mutation and deletion.This outcome probably result from demograohic history of rapid contraction that eliminated the diversity.
