本研究探討影響Streptococcus zooepidemicus ATCC 39920玻尿酸分子量的因素。在30至37°C之間,玻尿酸分子量隨培養溫度提高而略微增加,但影響並不顯著;在37°C下,玻尿酸分子量隨比生長速率提高而增加,比生長速率為0.1 h-1時,分子量為500 kD,比生長速率為0.58 h-1時,分子量約為900 kD。在對數生長中後期添加過氧化氫,並不能提高玻尿酸產率,而玻尿酸分子量則隨過氧化氫添加濃度提高而略微下降。研究結果顯示,改變鏈球菌比生長速率是獲得預期玻尿酸分子量較有效的方法。
The factors affecting the molecular weight (Mw) of the hyaluronic acid (HA) produced by Streptococcus zooepidemicus ATCC 39920 were examined. In the range of 30 to 37°C, the HA Mw increases slightly with the increase in cultivation temperature; however, the influence is not obvious. At 37°C, the HA Mw increases with the increase in the specific growth rate of cells. It was found that the HA Mw was around 500 and 900 kD when specific growth rate was 0.1 and 0.58 h-1, respectively The addition of H2O2 at the middle or late exponential growth phase is incapable of improving the production of HA. In addition, the HA Mw decreases as the increase in H2O2 concentration. The results indicate that the alteration in the specific growth rate is a more effective method to obtain the desired Mw of HA.
