羅勒,英名basil,臺灣俗稱為九層塔,為唇型花科Lamiaceae (Labiatae)羅勒屬(Ocimum)之植物。羅勒屬的品種分類,傳統上多以外表型態與精油成分加以分類,但這些性狀容易因為環境的因素而影響分類的結果。本試驗分析37個不同羅勒品種,除了外表性狀調查外,還運用ISSR分子標誌,分析不同品種在DNA層次上的差異,期能了解品種間的親源關係。ISSR試驗使用36個ISSR引子,經PCR增幅得到223個條帶,其中97%具有多型性。以UPGMA歸群方法,將37個品種分為4個群組:第一群包含了O. tenuiflorum;第二群包含了未知種名的香羅勒和藍香羅勒;第三群包含了具有檸檬香味的O. basillicum品種、O. americamum以及不具檸檬香味的O. gratissimum品種印度羅勒;第四群則全是O. basillicum的品種。本試驗證明利用ISSR能成功應用於羅勒品種分群上,有助於現有羅勒分類之研究,未來亦可利用此方法,將未歸群之品種歸屬至適當的分群當中,對於新品種的育成亦能提供相當之助益。
Basil (Ocimum spp.) is a member of Lamiaceae (Labiatae) family. In this study, morphological and ISSR markers were used to evaluate genetic relationship among thirty-seven basil. Basil could be divided into four groups based on ISSR marker analysis as below: (1) O. tenuiflorum included Sacred basil and Purple sacred basil; (2) O. sp. included Spice basil and Blue spice basil; (3) O. basillicum and O. americamum included four lemon-scented basil, O. gratissimum included Indian basil; (4) O. basillicum included tweenty-eight basil. The result of taxonomy by ISSR analysis was better than morphological analysis and was useful in discriminating among Ocimum spp.. In the future, molecular markers could be used in germplasm management of basil and distribute new unclassified basil to particular taxonomic group and reclassify basil to accurate classification.
