本研究介入餵糖不運動和禁食後不同運動時間,來觀察FOXO1對心肌的調節,AMPK活性和肝醣儲存,透過此研究希冀能了解運動和禁食效益對於心肌是否可以達到細胞週期減緩,進而使心肌細胞作用延長,達到延緩老化的影響 。研究方法對體重約250g的SD鼠50隻分配至5組,為控制組(Cf)、餵糖組(Cg)、運動一小時(1E)、運動三小時(3E)和運動三小時後休息1小時(ER),餵糖組給予每公斤體重1克葡萄糖(50%w/v)溶液,其他組均不補充。運動組分別給予單次運動一小時、三小時和三小時後休息一小時,和一組對照組。實驗結果發現,運動3小時組肝醣含量最少,運動3小時後休息一小時組肝醣含量最多。FOXO1磷酸化程度,餵糖組和禁食組沒有顯著差異,運動組以運動後3小時休息一小時磷酸化程度最多,但也沒有顯著差異。AMPK磷酸化程度,餵糖組和禁食組沒有顯著差異,運動組以運動3小時和運動3小時後休息一小時與控制組達顯著差異。本研究結論單次運動FOXO1對心肌調節沒有顯著影響,供能後無法立即減緩缺能的壓力;而運動後休息可使AMPK活化FOXO1進而調節能量信息,改善缺能環境,幫助細胞週期延長。
The purpose of the study is to know acute exercise of various times length with fasting condition and treatment glucose with no exercise would effect the cardiomuscle by observe ing FOXO1gene regulation, the activities of AMPK, and the storage of glycogen in rats. Through the study, it is hoped that the effects of exercise and fasting on decreasing cell cycle of cardiomuscle can be understood, thus finding ways to extending the cell cycle and slowing the effects of aging. 50 SD rats, each weighting approximately 250g, are assigned to 5 groups -- control (Cf), glucose feeding (Cg), one-hour exercise (1E), three-hour exercise (3E), and three-hour exercise with one-hour rest afterward (ER). Glucose feeding group is given 1 gram of glucose solution (50%w/v) per kilogram bodyweight. None of the other groups receive glucose.T he exercise groups are respectively given acute exercise for one hour, three hour, three hour with one hour of rest afterward, and a control group. The results show that the three-hour exercise group has the least amount of glycogen storage, while the three-hour exercise with one-hour rest afterward group has the most. As for the levels of phospholation of FOX01, there's no significant difference between the glucose feeding group and the fasting group. Among the exercise groups, the three-hour exercise with one-hour rest afterward group has the highest level of phosphoric acid, but without showing significant difference either. In terms of the levels of phospholation of AMPK, there's no significant difference between the glucose feeding group and the fasting group. Among the exercise groups, three-hour exercise (3E), and three-hour exercise with one-hour rest afterward (ER) significant with control. The research concludes acute exercise FOX01 has no significant effect to cardiomuscle, and adjustment that even with energy supply (glucose). We still find that rest after exercise could make AMPK activate FOXO1, then FOXO1 can regulate energy signal to help lack of energy condition and increasing cell cycle.
