寬尾鳳蝶屬(Agehana)的蝴蝶後翅具有不尋常的寬闊尾突,而且其內有兩條翅脈穿過,由於形態特徵與其他蝴蝶明顯不同,因此其分類歸群、親緣關係等尚未有明確定論,值得深入探討與釐清。目前一般認為這個屬有兩個物種,而台灣寬尾鳳蝶(A. maraho)被認為是台灣一個特有且瀕危的分類單元;另外一個物種是廣泛分佈於中國大陸南方的中華寬尾鳳蝶(A. elwesi)。本研究針對寬尾鳳蝶之屬級親緣關係及遺傳保育,分別進行三個部分的研究探討。
中華寬尾鳳蝶有一型的個體外表相似於台灣寬尾鳳蝶,被稱為中華寬尾鳳蝶白斑型,分佈地侷限於中國大陸西南地區一帶。此一白斑型與台灣寬尾鳳蝶以往並沒有從形態或分子方面作嚴格的檢視。第一部分的探討即針對雄性交尾器及CO I基因1531個鹼基對片段所做的基因序列檢測,結果顯示白斑型個體較近似於中華寬尾鳳蝶黑色型,與台灣寬尾鳳蝶關係較遠。此一結果支持白斑型個體僅是中華寬尾鳳蝶族群內之一「型」,而非亞種。
第二部份針對寬尾鳳蝶的屬級親緣關係進行探討,結果顯示寬尾鳳蝶屬的兩個分類單元彼此的分化時間極短且關係非常近緣,而與北美地區的Pterourus屬的鳳蝶結合成單系群。此一結果產生了一個極戲劇性的討論空間。本研究的初步建議暫時將寬尾鳳蝶屬處理成廣義鳳蝶屬 (Papilio sensu lato)的亞屬,或是併入Pterourus屬內。
先前以粒線體COI及COII序列分析,指出中華寬尾鳳蝶與台灣寬尾鳳蝶為兩個獨立種,但獨立種的分析差異小於0.5%。故欲利用完整粒線體DNA的定序分析,以了解寬尾鳳蝶兩獨立種間的分化時間。並以粒線體不同片段的序列分析,提供其它鱗翅目選用序列時的參考。第三部分的研究成功定出寬尾鳳蝶完整的粒線體基因體序列分別為 16093 bp(A. elwesi elwesi)、16095 bp(A. elwesi caveri) 及 16094 bp (A. maraho)。三個寬尾鳳蝶粒線體基因體的基因組成及排列方式與大部分的鱗翅目相同,僅在控制區(Control Region)與其它鱗翅目不同,具有大片段的重覆區(143bp及89bp)。三個基因體最大的變異區為cytb較一般常用的COI + COII基因的變異量多。整體的基因體變異點支持中華寬尾鳳蝶白斑型與中華寬尾鳳蝶黑色型的親緣關係較台灣寬尾鳳蝶為近。
