本實驗以肉毒桿菌神經毒素A型(Botox ®,Allergan Inc.,Irvine,OCA,USA)注射生長中大白鼠之咬肌與顳肌,造成咬肌與顳肌萎縮,降低咬合功能,藉此觀察其對於老鼠咬肌肌肉組織之影響。此實驗中所採取的研究方式為:選取60隻30天大的大鼠(Long-Evens),隨機分成四組,其中三組為實驗組、另外一組為控制組,依部位注射入大鼠。Group I:雙側咬肌注射肉毒桿菌神經毒素(顳肌注射入等量生理食鹽水);Group II:雙側顳肌注射肉毒桿菌神經毒素(咬肌注射入等量生理食鹽水);Group III:雙側咬肌和顳肌注射肉毒桿菌神經毒素;Group IV(control group):雙側咬肌和顳肌注射生理食鹽水。給予相同飲水與飼料,飼養45天後犧牲,取下咬肌及顳肌測量其重量,咬肌進行組織切片觀察肌纖維大小及肌纖維亞型(Type IIa, IIb, IIx)分布比例。本實驗得到的結論是,降低咀嚼肌的咬力,會使肌肉重量減低,並使肌纖維變細、並使肌纖維亞型發生轉換,則是使氧化能力較高的 type IIa fibers轉換為氧化能力較低的type IIb fibers。
<Objectives>In recent years, more and more patients receive Botulinum toxin A (BTXA) injection of masseter muscles to change their facial contour. The change is due to the reduction of masticatory function. Many past animal researches indicated that reduced masticatory function caused atrophy of muscle and morphological change in the growing period. But we still don’t understand exactly what happen on masseter muscle fiber after injection of BTXA. The purpose of this study is to clarify the changes of muscle fibers on histological level. <Materials and methods> 60 30-day-old rats were randomly divided into 4 groups. Each injection consists of using 25U/ml of BTXA divided according to : Group I-injection of masseter muscles. Group II-injection of temporalis muscles. Group III-injection of masseter and temporalis muscles. Group IV (control) –both muscles received 0.9% normal saline. After 45 days, the rats were perfused and sacrificed. The weight of masseter muscles were measured, and immunohistochemistry analysis of superficial masseter muscles was done. <Results and discussion> The size of muscle fibers were smaller in group 1,2,and 3 than control group. And there were a small percentage of type IIa fibers in study groups. <Conclusion> The reducing masticatory function may result in reduction of size of masseter muscle fibers and transition from type IIa fibers to type IIx or IIb fibers.
