青春期為性腺內分泌及配子生成的功能達到可生育的程度,具有繁衍能力的後代,在雌雄同體魚-黑鯛,其精卵共存、相互消長,及雄性先熟的特徵甚為獨特,故藉此作為實驗物種,探討精卵巢組織共存下,vgr、gpr54 及 gnrhr 基因在生殖腺性別發育及性轉變可能的作用及關聯。
本實驗中發現,vgr 確實顯著表現在卵巢組織,具有 age dependent 及 stage dependent 之特性,會高度表現在2+ 黑鯛,並至卵黃堆積後顯著下降;黑鯛經性類固醇 E2處理,卵細胞為初級卵母細胞,不易進入卵黃堆積時期而性轉變,可能原因之一應為 vgr 不易表現;vgr 表現和卵黃堆積及性轉變密切相關,精巢組織退化可使1+ 黑鯛提早至第二個繁殖季進行性轉變,vgr 亦確實顯著表現於卵黃堆積前期,但仍謹守 stage dependent 特性,故提早移除2+ 黑鯛精巢組織,卵細胞不會提早進入卵黃堆積,需直至繁殖季才可見卵黃堆積。
在 gpr54 及 gnrhr 確實在青春期早期表現,且無論1+ 或2+ 黑鯛,繁殖季或非繁殖季,在卵巢組織的表現量均高於精巢組織。透過 1+ 黑鯛 E2 處理以及精巢切除兩實驗,均不會影響 gpr54 及 gnrhr 在精巢及卵巢組織的表現量;利用2+ 黑鯛精巢切除以及單邊移除2+ 黑鯛生殖腺手術兩實驗可發現,gpr54 及 gnrhr均會在繁殖季 (Dec and Jan) 顯著表現,和性轉變密切相關。
最後根據結果推論,精巢組織退化為卵巢組織發育和性轉變的必要條件,vgr 需高度表現才可進行卵黃堆積及性轉變,gpr54 及 gnrhr 亦在性轉變扮演重要的角色。
