第二群過敏原是塵蟎主要過敏原的一種,分子量約為14~18 kDa 。多種塵蟎第二群過敏原皆具有高度的IgE反應普及率。Der p 2的IgE反應普及率高達80%~100%。本研究顯示Tyr p 2亦有相似結果。且專一性的IgE時常同時會和Der p 2以及Tyr p 2同時發生反應。由於Der p 2和Tyr p 2兩種過敏原之間具有高度的交叉反應,利用rTyr p 2可能可以預防Der p 2所引起的過敏反應的。因此本實驗再小鼠實驗動物模式中,利用重組Tyr p 2蛋白質(rTyr p 2)做為疫苗預防Der p 2所引起的過敏性氣喘反應。實驗中確認了Tyr p 2的IgE抗體結和特異區應位於C端的T87-Y107之間。而在疫苗療效的方面,rTyr p 2N端重組蛋白質(G16-N73)加上CpG寡核苷確實能有效抑止Der p 2致敏所引起的IgE抗體反應,也能產生防禦性抗體IgG2a,但是卻無法抑止IgG1抗體的上升。實驗也證明rTyr p 2N加上CpG寡核苷能夠藉由使免疫反應趨向Th1及Th2的平衡狀態,達到改善Der p 2所引起的氣管發炎反應的效果。
Group II allergens are a major mite allergen. The molecular weight of Group II allergens is about 14~18 kDa. The IgE binding prevalence of Der p 2 was as high as 80~100%. This result was similar to Tyr p 2 (group II allergen of storage mite, Tyrophagus putrescentiae). In our study, specific IgE react to both Der p 2 and Tyr p 2. Patients that specific allergic to Der p 2 or Tyr p 2 were rare. The inhibition test also reflected that Der p 2 and Tyr p 2 had high cross reactivity. So prevent Der p 2-induced allergic airway inflammation with Tyr p 2 maybe feacible. The purpose of this study was to use the epitope of rTyr p 2 as a peptide immunotherapy agent to prevent Der p 2 induced allergic airway inflammatory reactions in mice model. In E.coli expression system, deletion of the signal peptide can increase the yield and solubility of recombinant-protein.The IgE epitope of allergen is the major in inducing allergic response. Screening with allergic patients has shown that IgE epitope located between T87 -Y107 at C’terminal, but the IgG binding epitope was located at N’terminal. rTyr p 2N in conjunetion with CpG was used for nasal vaccination followed by Der p 2 immulization and airway challenge. The results showed that Der p 2 induced IgG and IgE production could be down regulated and airway imflamation could be inhibited. Since the IFN-γ+/CD4+ and IL-4+/CD4+ cells were reversed after vaccination, indicating that the anti-imflamation effect of Tyr p 2 N+CpG vaccination could be due to modulatory effect of Th1 and Th2 cells.
