腸病毒71型(Enterovirus 71, EV71)為一重要的人類重致病性病毒,在生物分類上是屬微小病毒科(picornaviridae)之腸病毒屬(enterovirus),為正單股(positive single strand)之RNA病毒。腸病毒71型在世界各地皆有流行,腸病毒71型在臨床上除手足口病(hand-foot-mouth disease)外,伴隨中樞神經系統疾病,例如心肌炎和腦炎,台灣地區於1998年爆發腸病毒71型大流行,造成405名孩童感染重症,78名孩童死亡,到目前為止台灣地區每年都有腸病毒流行,最主要的流行病株為腸病毒71型。目前針對腸病毒71型,尚無有效的藥物或疫苗可以預防或治療,有研究指出腸病毒71型感染宿主細胞的早期會引發細胞內PI3K/Akt、ERK、NFκB的活化,而這些訊息傳導途徑與調控細胞生長有關,病毒早期刺激細胞PI3K/Akt、ERK、NFκB的活化有利於病毒複製擴散並且持續性感染。先前本實驗室嘗試搜尋可以有效抑制腸病毒之天然食材,經多種樣品的測試後發現蘋果萃取液具有抑制腸病毒能力,因此決定進一步分析探討蘋果萃取液與其有效成分如何抑制腸病毒之機制做深入研究。我們的初步研究顯示蘋果萃取液與腸病毒71型作用可抑制病毒感染細胞所造成的PI3K/Akt、ERK、NFκB的活化,進而抑制病毒複製與持續性的感染。
Enterovirus 71 (EV71) is an important human pathogenic virus. Enterovirus 71 is a positive single strain RNA virus is a member of the Enterovirus genus of the Picornaviridae family. EV71 infection can cause hand-food-mouth disease (HFMD) is following the central nervous system (CNS), the disease, such as the myocarditis and encephalitis. In 1998, there was a large HFMD outbreak be heavy to cause 405 child infect with serious disease, 78 child death. EV71 is the most pathogenic virus in acute enterovirus complication in Taiwan till now. Effective medicine or vaccine that can be prevented or treated for EV71 is not yet available at present. Viruses have been known to subvert the anti-apoptotic pathways of the host cell in order to delay apoptosis. Previous studies have shown that EV71 can activation of the phosphatidylinositol 3-kinase (PI3K)/Akt trigger the anti-apoptosis pathway at the early-phase during infection and that Malus domestica can inhibits EV71 effectively. Thus, the virus-infected cells that signal transduction experiment, this is further purified extract can prevent the infection of cells caused by the activation of the PI3K/Akt.
