摘要
本論文應用二氧化氯對海水魚類所需初期動物性餌料生物(輪蟲、豐年蝦、短角異劍水蚤)的消毒抑菌效果與活存率,同時亦針對點帶石斑(Epinephelus coioides)魚授精卵消毒處理的抑菌效果及孵化率進行研究。此外利用 NNV(Nervous Necrosis Virus, NNV)病毒施打雞隻後所產生的蛋粉 IgY(Immunoglobulin Y)來滋養輪蟲、豐年蝦及短角異劍水蚤,進而投餵點帶石斑魚苗以進行育成率試驗,藉以提供一種被動性免疫(passive immunization)保護,減少病毒的感染,增加魚苗活存率。
餌料生物與石斑魚授精卵透過不同濃度二氧化氯(Chlorine dioxide)在不同時間內來進行消毒處理,實驗結果顯示豐年蝦及短角異劍水蚤各使用 4 ppm 二氧化氯處理 10 分鐘對豐年蝦、短角異劍水蚤的滅菌量為 100 % 且豐年蝦、短角異劍水蚤活存率亦為 100 %,輪蟲使用 4 ppm 二氧化氯處理 10 分鐘對輪蟲的滅菌量為99.2 %且輪蟲活存率為92.33 %;在石斑魚授精卵的消毒處理中以二氧化氯 2 ppm 處理 10 分鐘對石斑魚授精卵的滅菌量為99.2 %且輪蟲活存率為92.33 %可以大幅降低病原菌數量,同時不影響授精卵孵化率。
透過二氧化氯處理後孵化之魚苗,投餵已消毒並滋養抗 NNV IgY的餌料生物(輪蟲、豐年蝦及短角異劍水蚤)之育成實驗結果顯示,實驗組(投餵帶有抗神經壞死病毒 IgY 的蛋粉)的活存率為 5.79 %, 控制組的活存率為 4.36 %。攻毒實驗中亦顯示,注射 NNV 實驗組在注射後第 6 天的活存率為 43.33 %,注射 NNV 控制組在注射後第 6 天活存率為 0 %,結果顯示抗 NNV IgY 具有提高活存率的效果。

Abstract
During the culture of the larval stage of marine fish, live feed (such as: rotifer, copepoda, brine shrimp) is fundamental. The main purpose of this research was to investigate the effectiveness of chlorine dioxide in sterilizing the live feed and fertilized eggs of orange-spotted grouper (Epinephelus coioides). Chlorine dioxide was found to inhibit bacterial growth and increase the survival rate of live feed, at the same time increasing the hatching rate of orange-spotted grouper. In addition, Nervous Necrosis Virus (NNV) - IgY(Immunoglobulin Yolk), used to enrich live feed, acted as a passive immunization to viruses, so increasing the survival rate of grouper larvae.
Different concentrations of chlorine dioxide were used to sterilize live feed and fertilized eggs; results showed that a 10 minutes’ immersion bath at a concentration of 4 ppm was effective and did not result in harm to live feed. Similarly, a concentration of 2 ppm resulted in significant depletion of bacteria without affecting the hatching rate of the fertilized eggs.
Grouper larvae fed with sterilized, NNV-IgY-enrichment live feed, (the experimental group) survived at a rate of 5.79 %, while the control group (the feed of which was not enriched with NNV-IgY) survived at a rate of 4.36 %. Grouper larvae were challenged with a single dose of NNV pathogen, injected into the abdominal cavity. Survival rates were significantly higher in the experimental group than in the control group.
