地球上的生物由細胞所構成,細胞內帶有由DNA所形成的染色體之遺傳物質,所有的生命現象皆由DNA上所攜帶的密碼轉譯出的蛋白質來表現。DNA由五碳糖、磷酸根、嘌呤和嘧啶等小分子所組成,嘌呤和嘧啶為DNA上密碼的構成物質。DNA密碼的解譯,以Sanger sequencing酶鏈終止法為主要方法,搭配毛細管電泳為基礎的分析儀,成為目前主要使用的核酸定序分析儀,核酸定序成為分生實驗經常使用的項目之一。核酸定序若要成功,必須注意樣品和引子的純度及保存,還有實驗人員的操作純熟與否,這些因素都會影響核酸定序實驗的成功率。本篇即在探討有關核酸定序實驗成功與否的相關因素。
All creatures on earth are constructed with cells. Cells contain genetic materials which are made of DNA. All living things are characterized with the proteins expressed by codes on their DNA. DNA is a polymer of nucleotides. Each nucleotide is composed of pentose, phosphate, purine or pyrimidine. The genetic message is governed by the sequence of purine and pyrimidine. Sanger method which is specified by selective termination of DNA-polymerase, is a primary process to read the codes on DNA. The modern DNA-sequencing technique, an important tool in molecular biology, is a combination of Sanger method and capillary electrophoresis. Success in DNA-sequencing depends on the properties of sample and primer, and the skill of experimental operator. The factors affecting the results of DNA-sequencing are investigated in this work.
