論文名稱:以動物模式評估菊花木(Bauhinia championii)之鎮痛與抗發炎活性

菊花木(Bauhinia championii Benth.)為豆科羊蹄甲屬,是台灣民間常見之藥用植物,常用於治療跌打損傷。本研究探討菊花木抗氧化、鎮痛及抗發炎之活性探討,在化學抗氧化活性部分,以DPPH自由基清除力試驗、TEAC、還原力試驗、總多酚及總黃酮含量測定來評價抗氧化活性,在鎮痛活性方面,分別以醋酸扭體及福馬林試驗作為評估方法,醋酸扭體其方法為利用醋酸誘發小鼠因疼痛產生扭體反應,觀察其扭體次數來評估鎮痛效果,而福馬林試驗,其方法是利用福馬林誘導小鼠因疼痛造成舔足之反應,觀察其舔足時間來評估抑制疼痛之活性。
而在抗發炎方面,則以λ-carrageenan誘發小鼠足蹠腫脹,誘導五小時後,測其足蹠之丙二醛(MDA), NO, TNF-α, IL-1β, IL-6濃度及肝組織之抗氧化酵素(SOD, GPx, GRd)之活性含量變化。
實驗結果顯示菊花木具有抗氧化活性,並比同濃度之維他命C效果佳。而在鎮痛及抗發炎活性方面,能有效的抑制疼痛及發炎反應,其抑制發炎路徑為:經由抑制脂質過氧化作用進而提升體內抗氧化酵素GPx, GRd,而降低發炎反應。另一條路徑則為抑制細胞激素(TNF-α, IL-1β, IL-6)進而減少COX-2的合成,而使發炎反應降低。
綜合上述之結果,菊花木具有抗氧化、鎮痛及抗發炎活性,菊花木在台灣民間用藥上確有其使用之價值,且具有開發成新藥之潛力。

Bauhinia championii Benth. (Leguminosea) is a folk medicinal plant in Taiwan for the treatment of traumatic injury. In this research, we evaluated the antioxidant activities, analgesic effect, and anti-inflammatory effect of the stem of B. championii.
In the antioxidant study, we tested the methanol extract of B. championii. The total phenolic content, total flavonoid content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, trolox equivalent antioxidant capacity (TEAC), reducing power of B. championii were evaluated to explore its antioxidant activities. In the analgesic study, we tested methanol extract of B. championii by using animal models to observe acetic acid-induced writhing response and licking time in formalin test. The anti-inflammatory effect of B. championii was investigated by λ-carrageenan-induced paw edema in mice. We detected the activity of superoxide dismutase (SOD), glutathione peroxidase (GPx) and glutathione reductase (GRd) in the liver, the levels of interleukin-1β (IL-1β), interleukin-6(IL-6), tumor necrosis factor (TNF-α), malondialdehyde (MDA) and nitric oxide in the edema paw tissues.
Results showed, B. championii exhibited strong antioxidant activities in DPPH free radical scavenging effect, trolox equivalent antioxidant capacity (TEAC), and reducing power test. In the analgesic study, B. championii decreased the acetic acid-induced writhing response and licking time in the late phase of the formalin test. In the anti-inflammatory study, B. championii also decreased the paw edema due to λ-carrageenan administration. B. championii increased the activities of GPx and GRd in liver tissue and decreased MDA level in the edema paws at the 5th hour after λ-carrageenan-induced inflammation. B. championii also affected the levels of interleukin-1β, interleukin-6 and the level of TNF-α in the inflamed tissues.
In conclusion, B. championii has strong antioxidant activity, and has anti-inflammation and analgesic effects. It can be an important medicinal plant in Taiwan that is worth investigating for the development of new medicines in the future.
