黑色素的過度表現與累積,容易引起各類皮膚色素沉著的疾病。黑色素合成的控制受到酪胺酸酶、酪胺酸酶相關蛋白1、酪胺酸酶相關蛋白2及一些相關之轉錄因子的調控。[6]-生薑醇是生薑中的一種活性成分,具有抗發炎、抗氧化、抗癌、抗血管新生及抗胃潰瘍的藥理效性。為了要尋求新的皮膚美白試劑,[6]-生薑醇調節黑色素生成的效果和作用機轉被進一步的研究。在本研究中,我們利用分光光度計來測量[6]-生薑醇對蕈菇酪胺酸酶活性、細胞酪胺酸酶活性及細胞黑色素含量的影響;利用西方墨點法來測定[6]-生薑醇影響B16F10老鼠黑色素瘤細胞內酪胺酸酶及與黑色素生成相關之蛋白質的表現量。此外,我們還透過各種訊息傳遞抑制劑的作用來研究[6]-生薑醇抑制黑色素生成時可能的訊息傳遞路徑。為了解釋[6]-生薑醇抑制黑色素成的特性與其抗氧化活性之間的關係,我們也測定了[6]-生薑醇對B16F10細胞內活性氧分子(ROS)生成的影響。結果顯示,濃度25-100μM的[6]-生薑醇能依劑量增加有效地抑制老鼠黑色素瘤細胞內酪胺酸酶的活性及黑色素的含量,及有效地降低老鼠黑色素瘤細胞內ROS的表現程度。我們的結果指出[6]-生薑醇能有效地抑制B16F10細胞之黑色素生成及ROS的產生,因此[6]-生薑醇具有抑制黑色素生成的活性並結合清除ROS的能力,未來將可提供作為皮膚美白試劑。
Accumulation and overproduction of melanin leads to various skin hyperpigmentation disorders. Melanin synthesis process is regulated by tyrosinase, tyrosinase-related protein (TRP)-1 and -2, and some transcription factors. [6]-gingerol is an active component of ginger and shows pharmacological properties such as anti-inflammation, anti-oxidation, anti-carcinogenesis, anti-angiogenesis and anti-ulcerogenesis. To find a novel skin-whitening agent, the melanogeneis inhibitory effects and action mechanisms of [6]-gingerol were investigated.
In the present study, the effects of [6]-gingerol on mushroom tyrosinase activity, cellular tyrosinase activity and cellular melanin content were determined spectrophotometrically, expression of tyrosinase and related proteins in B16F10 murine melanoma cells were determined by Western blotting. Furthermore, the possible signaling pathway involved in [6]-gingerol-mediated depigmentation was investigated by specific inhibitors. To elucidate the relation between antimelanogenic property and antioxidant activity of [6]-gingerol, the effect of [6]-gingerol on reactive oxygen species (ROS) production in B16F10 murin melanoma cells was also assessed.
The results revealed that [6]-gingerol (25-100 μM) effectively suppress murine tyrosinase activity and the amount of melanin in a dose-dependent manner. Additionally, [6]-gingerol also effectively decrease intracellular ROS level in a dose-dependent pattern at the same dose range. To sum up, our results indicated that [6]-gingerol effectively inhibit melanogenesis and ROS production of B16F10 melanoma. We propose that [6]-gingerol exert antimelanogenic activity coupled with scavenging ROS property and would be a good skin whitening agent.
