肥胖是影響許多代謝異常的因子,造成脂質代謝異常及糖尿病等症狀,而糖尿病的高血糖現象所產生的氧化壓力為許多併發症的誘因。先前研究指出多醣體與多酚類可以有效降低高血糖現象及降低代謝症候群因子的發生,因此本研究主要探討虎奶菇(含豐富多醣體)、苦瓜及芭樂葉萃取物(含豐富多酚類)對於糖尿病大鼠降血糖及肥胖的影響。本研究分為三個部分:第一部分研究的目的,將STZ誘導的糖尿病大鼠飲食中添加虎奶菇胞外多醣體萃取物對其能量代謝的影響,探討其對肥胖的第2型糖尿病鼠減重的效果。結果發現,虎奶菇胞外多醣體萃取物可降低血脂和體重值,活化肝臟PPARα的表現量刺激脂肪利用減少堆積,並增加胰島素的分泌,達到減重的目的。第二部分的研究目的為給予虎奶菇胞外多醣體萃取物對抑制STZ誘導的糖尿病鼠高血糖之影響。結果發現,給予大鼠口服虎奶菇胞外多醣體萃取物能增加GPx活性和SOD值,降低了MDA的產生,並降低血糖,提高胰島素濃度達到改善糖尿病大鼠高血糖的情況。在第三部分的研究目的中,評估苦瓜和番石榴葉的多酚萃取物改善STZ誘導糖尿病老鼠高血糖效果。結果顯示,與非治療組比較,給予多酚萃取物組能有效降低血糖濃度,增加胰島素的分泌濃度,OGTT及ΔAUC的表現皆顯著降低。因此,從上述實驗結果,虎奶菇胞外多醣體萃取物和苦瓜、番石榴葉多酚萃取物具有潛力作為一種降血糖和減肥的功能性食品。
Obesity is a factor affecting many metabolic abnormalities. Previous studies indicated that polysaccharides and polyphenols reduced hyperglycemia and the incidence of metabolic syndrome factors. This study divided into three parts: The aim of first part study, we try to evaluate the effect of pleurotus tuber-regium extracellular polysaccharide extracts on energy metabolism with high fat diet and STZ induced obesity diabetic rat. The result found that pleurotus tuber-regium extracellular polysaccharide extracts reduced blood lipid values and the body weight, activation of the liver PPARα production, and increase insulin secretion to weight loss purposes. The aim of second part study, we examined pleurotus tuber-regium extracellular polysaccharide extracts for inhibitory effects against hyperglycemia on STZ induced diabetic rat. The result showed that oral administrated pleurotus tuber-regium extracellular polysaccharide extracts can increased GPx, SOD values and decreased the production of MDA, improved heart vascular oxidative stress, reduced blood glucose, enhance insulin level to conclude significant hypoglycemic effect in diabetic rats. The aim of third part study, we try to evaluate the effect of polyphenols extracts from bitter melon and guava leaf for hyperglycemia on STZ induced diabetic rat. The result showed that polyphenol extracts can effective in blood glucose level and increase insulin secretion levels in the group given polyphenol extracts and ΔAUC in the performance of OGTT were significantly lower than non-treatment group. Therefore, from the above experimental results, pleurotus tuber-regium extracellular polysaccharide extracts and bitter melon, guava leaf polyphenol extracts has potentials to used as a functional foods of hypoglycemia and weight lose.
