絲瓜組織中含有幾丁聚醣(Chitosan),而幾丁聚醣是目前頗受矚目的天然高分子材料,可廣泛應用在生物醫學、食品、農業、生技與藥品等領域。溫度影響植物的呼吸、光合作用與葉綠體中酶之活性,而改變了總碳水化合物含量。因此,本研究擬探討不同生長溫度與處理溫度對絲瓜幾丁聚醣之影響;同時評估其在植物生長與抑制病原菌之應用。

圓筒與稜角絲瓜均以30/27℃下生長最佳,也有較高的單株幾丁聚醣產量。當夜溫降低至25℃,幾丁聚醣含量增加,也提升單株幾丁聚醣產量。絲瓜組織中的幾丁聚醣含量隨著貯藏時間先增加而後減少。4℃貯藏至第15天,幾丁聚醣才低於採收當天的含量。絲瓜幾丁聚醣含量隨乾燥溫度提高而降低,而冷凍乾燥與35℃乾燥沒有明顯差異,表示絲瓜組織採收可貯放於4℃下1-3天,再以35℃乾燥處理可得到較高的幾丁聚醣含量。

幾丁聚醣能促進‘豐香’草莓生長與發育,亦能提升果實品質。在‘豐香’草莓上,100 mg L-1低分子量幾丁聚醣能效促進草莓的營養生長,20 mg L-1低分子量幾丁聚醣能提升‘豐香’草莓果實的品質。在抑菌上,絲瓜幾丁聚醣具有抑菌之效果,隨著濃度增加能明顯抑制甘藍黑斑病(Alternaria brassicae)菌絲的生長,處理至第七天,各組以濃度為1000 mg L-1皆有50%以上之抑菌率;在番茄細菌性軟腐病(Erwinia carotovora subsp. carotovora)上,96孔盤抗菌活性試驗顯示,三種幾丁聚醣皆有抑菌之效果。

Loofah tissue contains chitosan, a high polymer material that has attracted a lot of attention at present. It has been widely applied in biomedicine, food production, agriculture, biotechnology as well as medicine. High temperature can effect the respiration, photosynthesis, and activity of enzyme in the chloroplast, changing the total carbohydrate content.

The result in this study showed smooth and angled loofah had best growth at 30/27℃, and contain high chitosan content in them. When night temperature fell to 25℃, it increased the chitosan content and chitosan content per plant. Chitosan content following storage times first increases and then decreases. Storage to 15th day at 4℃, chitosan content was below the initial level of storage on the day. Increased drying temperature decreased chitosan content. Nevertheless, freeze-drying gets chitosan content equivalent to traditional 35°C. The result showed loofah tissue storage 1-3 day at 4℃, and use traditional 35°C can get higher chitosan content.

Chitosan stimulated growth and development, increased fruit yield and quality of strawberry ‘Toyonoka’. On strawberry ‘Toyonoka’, appropriate concentration and molecular weight of chitosan to stimulated vegetative growth was 100 mg L-1 Low molecular weight, and increased fruit quality was 20 mg L-1 Low molecular weight. The result showed Luffa-Chitosan has antifungal and antibacterial activity, and following increases concentration and increase supressive rate of Alternaria brassicae, and 1000 mg L-1 has over 50% supressive rate on 7th day. On tomato bacterial soft rot disease, 96 well plate antibacterial activity test showed all of group has antibacterial activity.
