本研究主要探討不同劑量馬黛茶葉對於 Streptozotocin (STZ) (50 mg/kg B.W.) 誘導後所形成的第 1 型糖尿病 Sprague-Dawley (SD) 大白鼠其醣代謝機制及對於脂質代謝之影響。實驗總共分成四組 (1) 正常組 (2) 糖尿病控制組 (3) 糖尿病組添加 1.5% 馬黛茶葉 (4) 糖尿病組添加 3% 馬黛茶葉,實驗共進行 4 週。結果發現,在脂肪重量方面,給予 3% 馬黛茶葉會顯著增加糖尿病大白鼠相對脂肪重量。此外,糖尿病大白鼠攝食 3% 馬黛茶葉 4 週後其飲水量及排尿量明顯降低。血漿生化值方面,3% 馬黛茶葉會明顯降低血糖濃度並增加胰島素濃度。在血脂濃度方面,3% 馬黛茶葉能有效降低糖尿病大白鼠血漿總膽固醇及三酸甘油酯濃度,1.5% 的馬黛茶葉則無影響。在肝臟方面,給予 3% 馬黛茶葉可以顯著降低糖尿病大白鼠肝臟總膽固醇及三酸甘油酯含量並降低脂肪酸合成酶 (fatty acid synthase, FAS) 之活性。在糞便脂質方面,給予 3% 馬黛茶葉會增加糖尿病大白鼠糞便中總膽固醇及三酸甘油酯的排出量。此外,1.3% 及 3% 馬黛茶葉會顯著增加肝臟 hexokinase 活性並降低 glucose-6-phophatase/hexokinase ratio,且具有劑量上的效應。3% 馬黛茶葉組其肝臟 glucose-6-phosphatase 活性及小腸雙醣酶 sucrase 活性明顯下降,肝臟肝醣的含量則有上升的趨勢。在蛋白表現量方面,1.5% 及 3% 馬黛茶葉會顯著增加肌肉中 p-Akt/Akt ratio且 3% 馬黛茶葉對 membrane Glut4/cytosol Glut4 ratio 有上升的趨勢。另外,測定大鼠血漿腫瘤壞死因子 (tumor necrosis factor, TNF-) 後,結果顯示 3% 馬黛茶葉組血漿 TNF-濃度顯著低於糖尿病控制組。此外,3% 馬黛茶葉也會降低糖尿病大白鼠血漿肌酸酐及尿素氮濃度,然而在血漿天門冬胺酸轉胺酶 (aspartate aminotransferase, AST) 及丙胺酸轉胺酶 (alanine aminotransferase, ALT),各組間則無統計差異。綜合以上,馬黛茶葉在調節糖尿病大白鼠醣類及脂質代謝具有改善的效果,且 3% 的效果更佳。
This study was designed to investigate the effects of mate tea leaves on carbohydrate and lipid metabolism in streptozotocin-induced type 1 diabetic rats. The experiments were divided into four groups: (1) normal group (2) type 1 diabetic control group (3) type 1 diabetic rats with 1.5% mate tea leaves (4) type 1 diabetic rats with 3% mate tea leaves. As the results, type 1 diabetic rats with 3% mate tea leaves were higher than type 1 diabetic group in relative adipose tissue weight. In addition, the drinking volume and urine volume of diabetic rats with 3% mate tea leaves were reduced compared with diabetic control group. In plasma biochemical values, we found that 3% mate tea leaves significantly lowered blood glucose and increased insulin concentration. In plasma lipid concentration, 3% mate tea leaves also significantly reduced the plasma total cholesterol and triglycerides concentrations in type 1 diabetic rats, however, it was no significant difference in 1.5% mate tea leaves group. Dietary supplementation with 3% mate tea leaves significantly lowered liver total cholesterol, triglyceride contents and fatty acid synthase activity and had a slightly increased in glycogen content. We also found that type 1 diabetic rats treated with 3% mate tea leaves increased fecal lipid excretion. 1.5% and 3% mate tea leaves significantly increased liver hexokinase activity and decreased glucose-6-phosphatase/hexokinase ratio. The glucose-6-phosphatase activity and intestinal sucrase activity in 3% mate tea leaves group were significantly inhibited. The protein expression levels of glucose transporter 4 were slightly increased in diabetic rats with 3% mate tea leaves. Moreover, after treatment with 3% mate tea leaves, we observed a recovery of the plasma tumor necrosis factor, blood urea nitrogen and creatinine concentrations. The results of this study suggested that mate tea leaves especially 3% may improve carbohydrate and lipid metabolism in type 1 diabetic rats.
