研究指出高血糖狀態會降低大鼠傷口膠原蛋白與細胞外基質含量。鱸魚 (Lateolabrax japonicas; LJ)、鮪魚 (Thunnus albacares; TA)、鮭魚 (Salmo salar Linnaeus; SL) 及鰻魚 (Anguilla japonica; AJ) 為臺灣飲食常見食用魚來源,傳統上亦認為食用魚為有助於手術後創傷修復之食材,因此實驗將探討鱸魚、鮪魚、鮭魚以及鰻魚對大鼠創傷修復之影響。分析食用魚經水萃後組成含量。大鼠隨機分配正常與高血糖兩組 (腹腔注射STZ 65 mg/kg BW 搭配菸鹼醯胺200 mg/kg BW 誘發大鼠高血糖),每隻大鼠背上製造6個全層皮膚切除傷口,分別塗抹去離子水 (DW)、速癒樂藥膏 (SO)、LJ、TA、SL及AJ處理,傷口連續塗抹6天於第7天進行犧牲。採集傷口組織分析,測定傷口收縮百分比、醣醛酸、六碳糖胺、氫脯胺酸、PDGF及TGF-β含量,評估食用魚對於大鼠創傷修復之影響。結果顯示LJ灰分、粗蛋白及無氮萃取物含量顯著高於其他魚種,SL與AJ粗脂肪含量顯著高於其他魚種。高血糖因子會顯著降低傷口醣醛酸、六碳糖胺及TGF-β含量;兩組傷口塗抹魚粉處理其醣醛酸、六碳糖胺、氫脯胺酸、、PDGF、TGF-β含量及傷口收縮百分比會顯著高於DW處理;LJ會增加高血糖組六碳糖胺、氫脯胺酸含量及傷口收縮百分比;TA會增加高血糖組醣醛酸、六碳糖胺、氫脯胺酸、PDGF含量及傷口收縮百分比;SL與AJ會增加高血糖組六碳糖胺、氫脯胺酸、TGF-β含量及傷口收縮百分比。綜合上述,食用魚可藉由不同修復機轉,達到促進傷口收縮之表現,且有助於高血糖狀態之創傷修復。
Previous studies have suggested that decreased production of collagen deposition and extracellular matrix contents all the impaired wound healing observed in hyperglycima rat. Sea bass (Lateolabrax japonicas; LJ), Yellowfin tuna (Thunnus albacares; TA), Atlantic salmon (Salmo salar linnaeus; SL), and Japanese eel (Anguilla japonica; AJ) is an ordinary fish source of daily diet in Taiwan. Also edible fishes are traditionally used for wound healing after surgery. The purpose of this study was to elucidate the effect of LJ, TA, SL, and AJ on wound healing in rats. Thus composition may be beneficial to the process of healing. Proximent analysis proceeded after the fish was cooked and freeze-dried. Sprague-Dawley rats were divided into normal (NR) and hyperglycemic (HG) group (induced by STZ 65 mg/kg and nicotinamide 200 mg/kg, intra-peritoneal). Each rat was created 6 wounds in full-thickness type extending up to the adipose tissue of back skin. After wounding, rats were treated at 24 hours with suile® ointment (SO), deionized water (DW), Sea bass (Lateolabrax japonicas; LJ), Yellowfin tuna (Thunnus albacares; TA), Atlantic salmon (Salmo salar linnaeus; SL), and Japanese eel (Anguilla japonica; AJ) for 6 days and sacrificed in the 7th day. The wound contraction was measured by using the epithelialization and granulation of tissue. Uronic acid, hexosamine, hydroxyproline, PDGF, and TGF-β were measured for describing the healing process. The results showed there ash, crude protein, amd nitrogen-free extract were significantly elevated in LJ compared to other fishes. The crude fat was significantly elevated in SL and AJ compared to other fishes. The results showed that hyperglycemia factor decreased significantly the concentration of uronic acid, hexosamine, and TGF-β. The results suggested a significantly increased in wound contraction, uronic acid, hexosamine, hydroxyproline, PDGF, and TGF-β contents with all fishes in NR and HG rats compared with DW. LJ significantly increased hexosamine, hydroxyproline, and wound contraction; TA significantly increased PDGF, uronic acid, hexosamine, hydroxyproline, and wound contraction; SL and AJ significantly increased TGF-β, hexosamine, hydroxyproline, and wound contraction. On the basis of these results, hyperglycemia dose did not favor the process of wound healing in proliferative phase. Fishes may be beneficial to the hyperglycemia wound contraction by mutiple wound healing mechanisms.
