運動員常補充營養增補劑,藉以改善及/或提升運動機能與表現,而中草藥是目前常被運動員使用之營養增補劑之ㄧ。冬蟲夏草(Cordyceps sinesis) 含有豐富的蟲草素(Cordycepin)、多醣體(Cordycepic polysaccharide)、腺核苷(Adenosine)、胺基酸等成分,有助於消除運動疲勞及增加運動表現,被廣泛作為營養增補劑使用,同時冬蟲夏草會增加血中睪固酮含量。基於睪固酮牽涉到運動表現與禁藥管制,因此長期使用冬蟲夏草對運動員之睪固酮變化值得注意。因此本論文探討以市售冬蟲夏草菌絲體當營養增補劑對不同訓練程度大鼠(S.D品系)之睪固酮濃度與代謝影響,並評估冬蟲夏草是否經由影響11β-HSD mRNA表現,因而造成大鼠睪固酮的變化。本實驗以酵素免疫分析儀測定大鼠體內睪固酮含量變化,亦測量肝臟CYP3A4酵素含量變化。最後應用Real Time-PCR儀器測定11β-HSD mRNA表現量。結果顯示市售冬蟲夏草菌絲體可提升睪固酮含量、抑制肝臟中CYP3A4酵素活性和增加睪丸中11β-HSD mRNA表現量。證實冬蟲夏草菌絲體於身體代謝機制中可有效幫助睪固酮含量提升。
Nutrition-enrich agents are frequently used by athletes for improvement and/or enhancement of sport function and performance. Herbal medicines have been applied by athletes as nutrition-enrich agents. Cordyceps sinesis, consisted of abundant cordycepin, cordycepic polysaccharide, adenosine and amino acids, has been wildly used as nutrition-enrich agent with beneficial effect of anti-fatigue and enhancement of sport performance. In addition, it has been reported to increase plasma testosterone concentration which is relative sport performance and doping control. Therefore, it is necessary to concern that the variations of testosterone concentration after long term uses of Cordyceps sinesis in athletes. The present study is to investigate the effect of commercial Cordyceps sinesis mycelium on the plasma concentration and metabolism of testosterone in exercised rat. In addition, the influence of Cordyceps sinesis mycelium on 11β-HSD mRNA is also evaluated. In this study, immunoassay was applied to measure the content of testosterone and CYP3A4 enzyme in the liver after administration of commercial Cordyceps sinesis mycelium product for 8 weeks. Finally, the expression of 11β-HSD mRNA was analyzed using a Real Time-PCR instrument. The results revealed that Cordyceps sinesis mycelium can enhance testosterone content, inhibit the activity of CYP3A4 enzyme in the liver and testis 11β-HSD mRNA expression. The results indicate that enhancement of testosterone content is associated with the inhibition of its metabolism.
