維生素E(Vitamin E)為一種脂溶性維生素,維生素E除了在食品的保存以及抗氧化之外,對於不同的疾病甚至於癌症皆能有效的預防、減輕症狀甚至於保護細胞免於受到傷害。甲氨蝶呤( Methotrexate, MTX)是一種非生物性疾病的抗風濕藥物,近代在其他疾病的治療上,也有許多新的研究成果展現出來。本研究使用MDA-MB-231細胞加入MTX模擬治療乳腺癌,並且加入維生素E,觀察是否會對於經MTX反應的MDA-MB-231細胞造成影響?我們使用臨床上較難治療的三陰性乳癌MDA-MB-231細胞,分別加入了0.1μM、10μM等濃度之MTX以及5μM的維生素E,待反應過後各自以MTT assay kit測其吸光值計算出細胞存活率。實驗結果發現MTX對於MDA-MB-231確實具有細胞毒性,在0.1μM組使細胞存活率下降10%,而濃度10μM組別的則顯著在第3天細胞存活率即下降35%;本實驗同時觀察維生素E與MTX合併使用後,發現維生素E對於MTX細胞毒性具有加乘作用,在MTX 0.1μM較低濃度組別也能有效使乳癌細胞死亡;我們使用自由基測定儀檢測細胞自由基含量,結果顯示加入MTX後,細胞內H2O2含量上升,且有濃度依賴性,其中MTX 10μM組的增加率達到500%,而加入維生素E發現,降低由MTX引起的細胞內自由基上升。本實驗證實,維生素E可使MTX在低劑量時即可有效使MDA-MB-231細胞死亡,且降低MTX造成的細胞自由基。期望未來可作為臨床癌症用藥一有效參考依據。
Vitamin E (Vitamin E) is a fat-soluble vitamin, vitamin E, and preserving food in addition to the antioxidant addition, even for different diseases are cancer can effectively prevent, alleviate symptoms even protect cells from harm. Methotrexate (MTX) is a non-biological disease modifying anti-rheumatic drugs, and modern times in the treatment of other diseases, there are many new research results show up. In this study, MDA-MB-231 breast cancer cells were added to MTX treatment simulation, and adding vitamin E, to observe whether the response to MTX by MDA-MB-231 cells were affected? We use clinically difficult to treat triple negative breast cancer MDA-MB-231 cells were added 10μM, 0.1μM of MTX. And 5μM concentrations of vitamin E, after each of the reaction to the MTT assay kit measured absorbance values to calculate the cell survival. Experimental results showed that MTX for MDA-MB-231 does have cytotoxic, cell viability at 0.1μM group fell by 10 percent, while the concentration of 10μM group decreased significantly in the third day of a fall of 35% cell viability; After this experiment observed MTX in combination with vitamin E, vitamin E was found to have a synergistic cytotoxic MTX role in, MTX 0.1μM lower concentration groups can effectively make breast cancer cell death; We used to detect free radicals in the radical analyzer, showed that after adding MTX, intracellular H2O2 content increased, and there is a concentration-dependent manner, where the rate of increase MTX 10μM group of up to 500%, while adding vitamin E was found to reduce radical rise caused by MTX within cells. In this study confirmed that vitamin E make MTX at low doses can be effective to MDA-MB-231 cell death, and reduces free radicals caused by MTX. We hope that the future can be used as an effective clinical cancer drug reference.
