豬生殖及呼吸綜合症廣泛常在於多數豬場中,主要引起離乳後豬隻之呼吸症狀及懷孕母豬之繁殖異常。本論文旨在評估PRRSFREE®次單位疫苗於母豬免疫補強反應,以及在一PRRS困擾之牧場評估PRRS疫苗及/或配合保育階段淨空之效益。試驗一於一具有懷孕後期流產疫情病史之牧場進行,於疫情爆發後,以PRRSFREE®疫苗於懷孕母豬進行免疫補強。試驗母豬於一劑或兩劑疫苗進行補強前及後,隨機採集血液進行免疫再活化反應評估,包括PRRSV特異性細胞內IFN-γ刺激試驗及PRRSV抗體監控。結果可見,母豬施打PRRSFREE®疫苗後,PRRSV特異性ELISA抗體顯著揚升,而施打兩劑疫苗之母豬所娩出仔豬,相較於一劑之組別亦具顯著較高力價之移行抗體;亦可見CD3+CD4+及CD3+CD8high之淋巴球數量增加。將周邊血液單核細胞以同源之野外活毒進行刺激後,可見CD4+及CD8+之IFN-γ分泌性細胞比例增加。更重要的是,PRRSFREE®免疫計畫於母豬應用後兩個月即可見流產疫情受到控制,顯示此些免疫反應似能反映在母豬之流產疫情控制及仔豬之保護上。試驗二則在一常年受到PRRS侵擾之牧場進行,於豬隻離乳後多見呼吸道疾病。在本試驗前,牧場內爆發新一波保育階段PRRS疫情,牧場管理人在隨即於未清場區開始兩PRRS免疫計畫的實行。結果可見保育階段豬隻死亡率明顯降低,然仍未達期望值。而部分豬隻於離乳後轉移至已淨空3個月之保育舍進行復養,並將其分為對照組及3個免疫計畫組。結果可見,施打疫苗之組別於保育階段死亡率及生長效能皆未見明顯改善。臨床呼吸症狀及治療頻率亦難見施打疫苗引起之顯著差異。此外,於豬隻6週齡前並無明顯之免疫後血清轉陽情形。然除此之外,飼養於淨空過豬舍之豬群於保育階段死亡率低於未清場過所飼養之豬群。綜合而論,除PRRS免疫策略,改善飼養管理策略之缺失及策略應用,包括保育舍清場再復養策略,對於高PRRSV感染壓力之牧場之疫情控制上為不可或缺的要素。

Porcine reproductive and respiratory syndrome (PRRS) characterized by respiratory distress on weaned piglets and reproductive failure has been endemic in most pig farms. The experiment was aimed to evaluate the boosting efficacy of the novel PRRSFREE® subunit vaccine on sows and the control efficacy of PRRS vaccine combined with or without nursery depopulation in a PRRS condemned farm. The first trial was conducted in a pig farm with abortion history following with a regular vaccination of PRRSFREE® subunit vaccine on pregnant sows. Blood samples randomly collected from the sows before and after one or twice vaccinations were submitted for recall antigen responses including PRRSV-specific intracellular IFN-γ production and antibody response. There was a significant increase in PRRSV-specific ELISA antibodies after sows vaccinated with PRRSFREE®. Maternal antibodies in suckling piglets were significantly higher in those piglets born from sows receiving 2 shots of vaccine compared to those with single vaccination. Both the number of lymphocyte subpopulation, especially CD3+CD4+ and CD3+CD8high and the frequency of CD4+ and CD8+ IFN-γ producing cells of peripheral blood mononuclear cells in response to live homogenous PRRSV stimulation were increased in sows vaccinated with PRRSFREE®. More importantly, the epidemic of abortion in the experimental farm ceased 2 months after the implementation of the vaccination program. The enhanced immunity may reflect on the abortion control and the protection of suckling piglets in the farm. The second trial was carried out on a PRRSV endemic farm that has been condemned with severe post-weaning respiratory disorders for several years. An epidemic PRRS occurred just before the experiment and two PRRS vaccination strategies had been tested in non-depopulated nursery units by owner’s discretion which showed marked decreased in nursery mortality, but not reached the expectant value. In contrast, some weaned pigs were transferred into a depopulated nursery house for the vaccine control study including one control and 3 different vaccine program groups. Neither nursery mortality reduction nor growth performance improvement could be found among those vaccine groups performed in the depopulated nursery unit. There were also no significant difference of clinical respiratory scores and frequencies of treatment among control and vaccinated groups. Additionally, no obvious PRRSV seroconversion of vaccinated pigs could be found before 6 weeks of age. Besides evaluation of vaccine efficacy, non-vaccinated pigs raised on depopulated house showed obviously decreased nursery mortality compared to weanling piglets raised on non-depopulation house. Taken together, besides PRRSV vaccination strategies, improvement of environmental and managerial detrimental factors, including nursery depopulation strategy, is required for PRRSV control in the farm with high infectious pressure.
