本研究使用報導基因重組酵母菌試驗法檢測臺灣西部五條河川 (北港溪、鹽水溪、二仁溪、阿公店溪、高屏溪)及高雄港區中底泥之類(抗)雄激素、類(抗)鹽皮質激素及類(抗)雌激素活性,並配合氣相與液相層析質譜儀分析底泥中之內分泌干擾物質。生物試驗法結果顯示,河川與高雄港底泥中並不具有類雄激素、類雌激素活性,類鹽皮質激素活性僅於二仁溪中的兩個採樣點中測得,而所有採樣點均有測出抗雄激素活性 (〈69.1-6224.4 FLU-EQ μg/g)、抗鹽皮質激素活性 (〈20.8-16506.6 SPL-EQ ng/g)及抗雌激素活性 (〈3.1-284.3 OHT-EQ μg/g),其中以二仁溪、阿公店溪與高雄港區的採樣點測得之抗雄激素、抗鹽皮質激素及抗雌激素活性最為顯著。這些河川採樣點都位於受民生、工業與畜牧廢水污染嚴重之地區,高雄港區則飽受重工業及海運交通業的污染。儀器分析結果顯示,底泥中常檢出多種會對人體及生物造成內分泌干擾、致癌性或生長發育影響的污染物質,包括多環芳香烴 (534-18369 ng/g)、多溴聯苯醚 (1.8-217.0 ng/g)、天然雌激素 (18.6-91.0 ng/g)、雙酚A (ND-1223.5 ng/g)、壬基酚 (ND-5458.3 ng/g)、三氯沙 (ND-829.7 ng/g)及三氯卡班 (ND-739.3 ng/g)等。比較生物試驗法與儀器分析換算之當量濃度結果發現,生物試驗法測出之內分泌干擾活性較高,顯示底泥樣本中可能具有非本研究儀器分析之對象物質,而這些物質可能是貢獻內分泌干擾活性的主要物質。
Many kinds of endocrine disrupting chemicals (EDCs) have been used in our daily lives. EDCs are able to interfere with the endocrine system, thus probably cause adverse effects on the organisms. In this study, recombinant yeast bioassays were used to detect androgen receptor (AR), estrogen receptor (ER) and mineralocorticoid receptor (MR) disrupting activities in sediment samples from Taiwanese rivers and Kaohsiung Harbor. In addition, sediment samples were also analyzed by gas and liquid chromatography mass spectrometry (GC/MS, LC-MS/MS) to quantify the concentrations of target polycyclic aromatic hydrocarbons (PAHs), polybrominated diphenyl ethers (PBDEs) and EDCs. Bioassay results revealed that AR, MR, ER antagonist activities were detected in sediment samples, whereas no agonist activity was found. Instrumental analysis showed that the most often detected compounds were 2-4 rings PAHs, decaBDE, bisphenol A, nonylphenol, triclosan, and triclocarban. However, they were not the leading contributors to the AR, MR, and ER antagonist potencies observed in the river sediment samples. Compared to other studies, PAH concentrations in the harbor sediments were generally higher than river sediments. In future work, we will try to identify the unknown endocrine disruptors in sediment samples
