白點症病毒(white spot syndrome virus, WSSV)主要以海洋甲殼類為宿主,並且對對蝦類有極高的致死率,自1992年蝦白點症爆發以來,造成水產養殖業的經濟損失,然而目前仍缺乏有效的治療方式。白點症病毒於感染宿主後首先藉由宿主的轉錄因子協助表現其極早期基因,這些基因包含病毒的轉錄調控因子,因此在病毒的生活史中是連結宿主與病毒調控機制的樞紐。極早期蛋白質WSSV108、WSSV126 (IE1)、WSSV136及WSSV156曾被報導具有轉錄調節活性,但目前對於白點症病毒自身基因的轉錄調控機制仍了解不多。本研究室先前研究發現白點症病毒極早期蛋白質WSSV108能活化wssv304及其他的病毒基因,驗證了WSSV108在細胞中具有調控轉錄的能力,而本研究透過免疫螢光染色發現WSSV108會聚集於細胞核中,確實可能參與轉錄作用。本研究也透過GST pull-down及冷光報導分析,發現WSSV108可調控其自身的啟動子,也會與白蝦(Litopenaeus vannamei)轉錄因子LvYY1的N端區域結合,並共同促進wssv108啟動子的轉錄,顯示wssv108的表現存在正回饋的機制,利於病毒快速表現WSSV108並促進其下游基因表現。而透過RNA干擾抑制病毒wssv108表現則導致病毒複製能力大幅減弱,並顯著提升了宿主受病毒感染後的存活率,顯示WSSV108對於白點症病毒是複製與致病過程中重要的蛋白質。總結上述結果,本研究發現WSSV108在白點症病毒的生活史扮演重要的角色,可能透過與宿主的LvYY1共同調控病毒的基因表現。
