研究目的:本篇研究台灣自然停經婦女的BRSK1基因-SNP rs12611091、rs1172822、rs7246479與自然停經年齡之間的相關性。
材料方法:研究利用臺灣人體生物資料庫(Taiwan Biobank),探討台灣婦女自然停經年齡及BRSK1基因SNP rs12611091、SNP rs1172822及SNP rs7246479。並進一步探討有無運動,對其BRSK1基因與停經年齡的關係。研究變項包含SNP rs12611091、SNP rs1172822、SNP rs7246479、身體質量指數BMI、抽菸習慣、飲酒習慣、運動習慣、初經年齡、哺乳經驗,並以一般線性迴歸分析BRSK1基因型與自然停經年齡的關係。
研究結果:研究對象共計3131人進行分析,平均停經年齡為50.78歲(標準差3.63歲)。SNP rs12611091,TC型β值為-0.31(P值0.018),顯示帶有1個變異其停經年齡會提早0.13年。TT型β值為-0.53(P值0.043),顯示帶有2個變異其停經年齡會提早0.53年;SNP rs1172822,TC型β值為-0.38(P值0.024),顯示帶有1個變異其停經年齡會提早0.38年。而TT型變異沒有達顯著意義;SNP rs7246479,GT型β值為-0.42(P值0.003),顯示帶有1個變異其停經年齡會提早0.42年。而GG型變異就沒有達顯著意義。進一步運動分層下,在沒有規律運動組中,BRSK1 rs12611091,TC型β值為-0.57(P值0.005)、TT型β值為-1.15(P值0.005),顯示帶有次要等位基因(T)會顯著提早停經。但在有規律運動組中,則未發現相關性;BRSK1 rs1172822在運動分層後皆未發現顯著性;BRSK1 rs7246479在未規律運動組中,帶有GT型變異會使停經年齡提早0.61年且達統計顯著(P值0.005) 而有兩個變異GG型會提早0.48年,但未達統計顯著(P值0.358)。
結論:台灣女性帶有BRSK1基因 rs12611091基因型變異,可能與自然停經年齡提早相關,而且變異愈多愈早停經。對有運動的人而言,這些基因型的變異並沒有提早停經的趨勢,但沒有運動的人當中,rs12611091變異的人會提早停經的現象更明顯,而且變異愈多愈早提經。所以更加帶有此rs12611091基因型應鼓勵多加運動。
Objectives: In this study, we investigated the association between BRSK1 gene-SNP rs12611091, rs1172822, rs7246479 and age at natural menopause in Taiwan.
Methods: The study used the Taiwan Biobank to explore the age at natural menopause and the BRSK1 gene SNP rs12611091, SNP rs1172822 and SNP rs7246479. Further exercise stratification, the relationship between BRSK1 gene and menopause. The study variables included SNP rs12611091, SNP rs1172822, SNP rs7246479, body mass index BMI, smoking habits, drinking habits, exercise habits, age at menstruation, breastfeeding experience, and the relationship between BRSK1 genotype and age at natural menopause by general linear regression analysis.
Results: A total of 3,131 subjects were analyzed, and the average age of menopause was 50.78 years (±3.63 years). SNP rs12611091 showed significant association with TC (β=-0.31, P= 0.018) and TT genotypes (β=-0.53, P=0.043).SNP rs1172822 showed significant association with TC genotypes (β=-0.38, P=0.024) but TT genotypes variation did not found significant; SNP rs7246479 showed significant association with GT genotypes (β=-0.42, P=0.003) but GG genotypes mutation did not found significant; Under further exercise stratification, SNP rs12611091 showed significant association with TC genotypes (β=-0.57,P=0.005) and TT genotypes
(β=-1.15, P=0.005) in the irregular regular exercise group. However, in the regular exercise group, no correlation was found; SNP rs1172822 was not shown significant after exercise stratification; SNP rs7246479 in the irregular exercise group, show significant with GT genotypes (β=-0.61, P = 0.005), but not significant with GG genotypes (β=-0.48, P = 0.358).
Conclusion: Taiwanese women with BRSK1 rs12611091 genotype variation may be associated with early age at natural menopause. If the location of the Allele variation is more, the age of menopause will be earlier. People with regular exercise, genotype variation do not have a tendency to early menopause. However, among those who do not have regular exercise, the phenomenon of early cessation of rs12611091 variation is more obvious. And the more allele variation, the menopause will be early.
