Introduction: Branched-chain amino acids (BCAA) and citrulline are common supplementation in sports. BCAA may reduce the cerebral uptake of free tryptophan, resulting in lower serotonin synthesis in the brain. The lower cerebral serotonin levels may reduce central fatigue. However, BCAA supplementation could lead to increases in plasma ammonia during exercise due to the elevated BCAA oxidation. Ammonia is known to induce central fatigue, which could offset the potential ergogenic effect of BCAA. Citrulline could remove plasma ammonia by increasing urea cycle and vasodilation. Therefore, the combined supplementation of BCAA and citrulline may increase exercise performance by reducing central fatigue. It has been suggested central fatigue contributes to the decline in performance in resistance training. Purpose: The purpose of this study is to investigate the effect of BCAA and citrulline supplementation on the rate of force development in the lower limbs after high-intensity resistance exercise. Method: Ten healthy male collegiate students with at least 6 months of resistance training experience were recruited for this study. Each subject completed both AA and PLA trials. In the AA trial, the subject consumed BCAA 0.1 g/kg body weight and citrulline 0.025 g/kg body weight. In the PLA trial, the subject consumed placebo. After the supplementation, the subjects underwent 5 sets of 5 repetitions of squat at 85% 1 RM. Isometric mid-thigh pull test was performed before, immediately after, and 24 hr after the squat training. The peak force and rate of force development at 50, 90, 150, and 200 ms were measured. Blood samples were collected before breakfast and before, after, and 24 hr after the squat training. Plasma levels of lactate, glucose, uric acid, ammonia, urea, and creatine kinase were measured. Results: The peak force and rate of force development in all time points in the isometric mid-thigh pull test in were not significantly different between the 2 trials. All blood parameters were similar between the 2 trials in all time points. Plasma ammonia concentrations before, immediately after, and 24 hr after the training were significantly lower than the baseline. On the other hand, plasma ammonia concentrations were similar in all time points in the AA trial. Conclusions: Supplementation of BCAA and citrulline has no effect on muscular performance in lower limbs after high-intensity resistance training in healthy male subjects with resistance exercise experience. The blood biochemical parameters were also similar between the supplement and placebo trials. This supplementation method has no effect on resistance exercise performance.
