牛樟芝 (Antrodia Cinnamomea)是台灣特有的稀有且昂貴的藥用真菌,經近年來科學研究發現其成份含有三萜類化合物、多醣體、超氧歧化物、免疫蛋白、維生素類、麥角醇類、核酸、氨基酸等等。許多的文獻指出牛樟芝中的三萜類化合物為抗癌的主要成分之一,而樟芝酸K (Antcin K) 即是從牛樟芝中分離出來的一種三萜類化合物 (triterpenoids)。本篇論文以研究Antcin K處理肺癌A549細胞後,分別進行MTT assay、Wound healing assay及Boyden chamber invasion assay等實驗,來測試Antcin K 對肺癌細胞A549的細胞存活率、爬行及侵襲能力的影響。首先在MTT assay的實驗中,隨著Antcin K的濃度逐漸增高,細胞的存活率依次降低;其次在Wound healing assay與Boyden chamber invasion assay的實驗中,亦發現隨著Antcin K濃度的增高,A549細胞爬行及侵襲能力亦受到抑制。這些研究數據顯示Antcin K對肺癌A549細胞有抑制作用,Antcin K可能會是治療肺癌有潛力的藥物,也期待著Antcin K能在未來治療肺癌臨床應用上得到進一步的突破。
Antrodia Cinnamomea is a rare and expensive medicinal fungus unique in Taiwan. In recent years, scientific research has found that its ingredients contain triterpenoids, polysaccharides, superoxide dismutants, immune proteins, vitamins, ergots, nucleic acids and amino acids, etc.Many documents point out that the triterpenoids in Antrodia cinnamomea are the anticancer components and Antcin K is a triterpenoids isolated from Antrodia cinnamomea. This paper investigates the effects of Antcin K on lung cancer A549 cells by MTT assay, Wound healing assay and Boyden chamber invasion assay to verify the effect of Antcin K on cell proliferation,migration and invasion. First, in the MTT assay experiment, as the concentration of Antcin K gradually increased, the cell survival rate decreased in turn; secondly, in the experiments of Wound healing assay and Boyden chamber invasion assay, it was also found that as the concentration of Antcin K increased, A549 cell migration and invasion ability is also suppressed. These data show that Antcin K has an inhibitory effect on lung cancer A549 cells. Antcin K may be a potential drug for the treatment of lung cancer. It is also expected that Antcin K can achieve further breakthroughs in the clinical application of lung cancer in the future.
