黑米 (Oryza sativa L.) 是色素米的一種,它含有多種營養和生物活性成分,包括必需氨基酸、功能性脂質、膳食纖維、維生素、礦物質、花青素、酚類化合物、γ-谷維素、生育酚、生育三烯酚、植物甾醇和植酸。許多研究表明,黑米具有抗氧化、抗炎、抗糖尿病、抗癌和降血脂等作用,但黑米之免疫調節效能仍不清楚。為了研究日常補充黑米對免疫調節的影響,本研究對養生黑米粥 (HBRP) 及養生台稉9號糙米粥 (HTK9P) 的生物活性物質 (總花青素、總類黃酮及總多酚化合物) 進行定量分析,並以BALB/c小鼠探討養生米粥對免疫調節的影響。實驗方法: 使用呈色法對總花青素、類黃酮及多酚化合物進行定量分析,小鼠犧牲後採集血清檢測血脂、血糖、TNF-α、IL-6、IL-10、IgG及IgM 之含量,並測量心、肝、脾、肺、腎及脂肪之重量,將脾臟細胞懸浮液加入LPS共同培養後,取上清液檢測 TNF-α、IFN-γ、IL-1β、IL-6及IL-10 的含量,使用MTT法分析養生米粥對脾臟淋巴細胞增生能力之影響。實驗結果顯示,HBRP的總花青素、總類黃酮及總多酚化合物含量皆顯著高於HTK9P,HBRP組小鼠之血清三酸甘油脂濃度顯著低於控制組及HTK9P組,HBRP及HTK9P皆可抑制小鼠脾臟分泌促發炎性細胞激素 (TNF-α及IL-6),HBRP可提高小鼠血清IgM含量並降低血清TNF-α 含量。由上述結果推論HBRP可透過抑制小鼠血清 TNF-α、脾臟 TNF-α 與脾臟 IL-6 的生成,以及提高血清 IgM 濃度來達到調節免疫的效果。

Black rice (Oryza sativa L.) is a variety of pigmented rice. It contains numerous nutritional and bioactive components, including essential amino acids, functional lipids, dietary fibre, vitamins, minerals, anthocyanins, phenolic compounds, γ-oryzanols, tocopherols, tocotrienols, phytosterols and phytic acid. Many studies shows that black rice has anti-oxidation, anti-inflammation, anti-diabetes, anti-cancer and hypolipidemia effects, etc. However, the role of black rice in immunoregulation is still unclear. To investigate the immunoregulating effects, the amount of total anthocyanins, total flavonoids and total polyphenolic compounds in health black rice porridge (HBRP) and health Taiken 9 (brown rice) porridge (HTK9P) were quantified using the colorimetric methods. In vivo study, 60 male BALB/c mice (5-week-old) were separated into 5 groups and HBRP/HTK9P were administrated (oral gavage) in different dosages for 28 days. After euthanasia, serum were collected for analyses including blood lipid, blood sugar, immunoglobulin (IgG and IgM) and cytokines (TNF-α, IL-6, IL-10) levels. Cytokines (TNF-α, IFN-γ, IL-1β, IL-6 and IL-10) levels and proliferational rate of splenocytes. The biologically active substance levels in HBRP were significantly higher than HTK9P, especially total anthocyanins (P<0.001). The levels of triglycerides and TNF-α in serum were significantly lower in the HBRP groups and both HBRP and HTK9P inhibited the secretion of IL-6 and TNF-α in spleen. HBRP increases the IgM level. The results suggested that black rice
may ¬affect immune response by regulating the levels of TNF-α and IgM in the serum of mice.
