玉山杜鵑複合群(Rhododendron pseudochrysanthum complex) [包括玉山杜鵑(R. pseudochrysanthum Hayata)、森氏杜鵑(R. morii Hayata) 、紅星杜鵑(R. rubropunctatum Hayata)及南湖杜鵑(R. hyperythrum Hayata)]的分類地位及種的界限時有爭論,本研究以葉綠體DNA atpB-rbcL noncoding spacer及細胞核DNA ITS 非轉錄區間重建玉山杜鵑複合群親緣地理關係,結果顯示玉山杜鵑、森氏杜鵑及紅星杜鵑應合併為同種,南湖杜鵑和玉山杜鵑近緣且為不同種,其中玉山杜鵑種化時間估算約在1.4~2.1百萬年前,應在恭茲冰期(Gunz;距今約1.2~2百萬年前)遷入台灣,並分別進入玉山山脈及中央山脈,推測約在1.22~1.83百萬年前因板塊擠壓導致山脈隆起造成兩個地理區族群的隔離,進而開始分化形成玉山山脈分支族群及中央山脈分支族群並擁有高度遺傳多樣性,根據玉山山脈分支族群累積較高遺傳歧異度,推測此族群應為久遠演化歷史之族群或是來自兩個具有高度分歧之族群二次接觸後譜系混雜;根據Tajima’D為顯著負值、族群變異分佈測驗(mismatch)中葉綠體DNA及細胞核DNA均呈現單峰狀態以及在網狀親緣樹狀圖顯現星狀圖放射狀等種種證據顯示中央山脈族群在其演化歷史曾發生過長期或嚴重的瓶頸效應,由少數先驅者祖先型快速族群擴張造成核苷酸歧異度偏低;南湖杜鵑種化時間約在0.53~0.79百萬年前,約在明德冰期(距今約0.73~1.0百萬年前)進入中央山脈北部,冰河退卻後因地理隔離而開始分化,隨著分隔時間增加種化為新種;玉山杜鵑和南湖杜鵑雖在葉綠體DNA顯現分化,但在細胞核DNA彼此混雜,顯現兩分類群間曾有雜交事件產生。由於花粉和種子傳播能力不同造成不同分子標記所顯現之族群動態不一致,玉山杜鵑因顯現種子傳播歷史之葉綠體DNA受到傳播距離影響而產生地區性分化,但顯現花粉傳播歷史之細胞核DNA則可藉由長距離傳播、不受地理隔離而無顯著分化。
In this study, we examined the taxonomic status and conservation genetics of the Rhododendron pseudochrysanthum complex, including R. pseudochrysanthum Hayata, R. morii Hayata, R. rubropunctatum Hayata, and R. hyperythrum Hayata. Based on the maternally inherited cpDNA and biparental inherited nrDNA variation, the reconstructed phylogeny reveals a conspecific relationship among R. pseudochrysanthum, R. morii, and R. rubropunctatum Hayata. However, R. hyperythrum Hayata should recognize as an independent taxon. According to the molecular clock, the speciation time of R. pseudochrysanthum was about 1.4~2.1 million years ago, and the divergent time between the populations of different Mountain Range of R. pseudochrysanthum was about 1.22~1.83 million years ago. The cpDNA genealogy of R. pseudochrysanthum reveals that the populations between Central Mountain Range and Yushan Mountain Range possess high levels of genetic diversity. Significantly negative Tajima’D values of cpDNA, star-like distribution in NCA of cpDNA, and the single peak of mismatch of cpDNA and nrDNA reveal that the populations of the Central Mountain Range have been experienced rapid population expansion with low values of nucleotide diversity. High values of nucleotide diversity and haplotype diversity in the populations of Yushan Mountain Range may attributes to secondary contact between differentiated allopatric lineages or to a long evolutionary history of a large stable populations. The splitting of R. hyperythrum from R. pseudochrysanthum was dated to 0.53~0.79 million years before present and then entered to the north parts of the Central Mountain Range. According to the ITS analysis, hybridization had happened between R. pseudochrysanthum and R. hyperythrum. The different spread of cpDNA and nrDNA may attributed to differentiate population lineages.
