背景與目的:停經婦女因雌激素分泌減少,氧化壓力增加,使血管內皮細胞結構及功能受損,導致血管病變,增加高血壓相關心血管疾病發生的機率,因此停經後婦女如何預防及控制高血壓是很重要的。本研究主要目的為探討八週運動及原兒茶酸介入對停經合併高血壓之血管舒張反應和抗氧化功能的影響,並進一步研究其生理機制。方法:本研究以雌性自發性高血壓大鼠(spontaneously hypertensive rat, SHR)為動物模式,隨機分為假手術組(SHR-sham)、卵巢切除組(SHR-O)、和卵巢切除合併運動和原兒茶酸補充組(SHR-OEP),並以週齡相符之正常血壓鼠(Wistar-Kyoto rat, WKY)作為對照組。運動訓練為進行跑步機訓練,每日六十分鐘,每週五天,為期八週。原兒茶酸補充則以每天每公斤體重 200毫克之原兒茶酸加入飲用水中,餵食八週。八週介入結束後分別測量四組之血管舒張功能和血管相關蛋白質表現量,並檢測肝臟脂質過氧化物。結果:本研究發現與假手術組和卵巢切除組比較,經八週運動訓練及原兒茶酸合併介入後結果顯示:一、顯著增加胰島素和類胰島素生長因子-1調節之血管舒張功能 (p < 0.05) ; 二、顯著增加血管中內皮細胞一氧化氮合成酶、胰島素受器和類胰島素生長因子-1受器之蛋白質表現量(p < 0.05); 三、肝臟脂質過氧化物含量分析中,發現經八週介入後SHR-OEP組顯著低於SHR-O組。結論:本研究發現八週運動及原兒茶酸介入能有效改善停經合併高血壓之血管舒張功能。
Background and purpose: Menopause-related estrogen deficiency and oxidative stress increase the risk of hypertension-related cardiovascular disease in postmenopausal women. Thus, the prevention and control of hypertension is an important issue for postmenopausal women. The purpose of this study was to investigate the effects of 8-weeks exercise training combined with protocatechuic acid (PCA) supplementation on vasorelaxation and antioxidation in postmenopause with hypertension. Methods: The female spontaneously hypertensive rats (SHR) were used as the animal model. They were randomly divided into three groups: the sham operation (SHR-sham), the ovariectomy (SHR-O), and the ovariectomy with exercise training and PCA supplementation (SHR-OEP). The Wistar-Kyoto (WKY) rats were used as the normotensive control group. Exercise training was conducted by the treadmill for 1 hr/day, 5 days/week for 8 weeks. PCA was supplemented with PCA solution (200 mg/kg/day) for 8 weeks. At the end of experiments, the vasorelaxation, vascular protein expression, and liver oxidation were measured among four groups. Results: Compared with the SHR-sham and SHR-O groups, the 8-week intervention significantly improved the insulin-induced and IGF-1-induced vasorelaxation (p < 0.05), it significantly enhanced protein expressions of eNOS, IR, and IGF-1 R(p < 0.05). Moreover, liver lipid oxidation was significantly reduced in the SHR-OEP group compared with the SHR-O group. Conclusion: This study suggested that 8-week exercise and PCA intervention significantly improved vasorelaxation in postmenopause with hypertension.
