Angelicae Radix and Ligustici are two commonly used Chinese herbal drugs. They have similar constituents and pharmacological activities, and are used together in many Chinese herbal preparations. It is found that the drugs possess sedative, antispasmodic, antiinflammatory and blood level decreasive effects and contain essential oils, amino acids, fatty acids, orangic acids and carbohydrates. In this study, we used orangic acids and essential oils as indexing standard to investigate the differences between samples of different growing times and different parts from the plants of Angelica acutiloba Kitagawa grown in Taiwain by high-performance liquid chromatography. The quality of commercial Angelicae Radix and Ligustici Rhizoma has also been studied by this method.The analytical data showed that the highest contents of the individual components were that ligustilide in flowers(6.98 mg/ g) and fruit(8.49 mg/g), butylidenephthalide in fruit(0.59 mg/g) and ferulic acid in leaves(0.25 mg/g) before blooming. The contents of commercial Angelicae Radix were as follows: nicotinic acid 0-0.57 mg/g, phthalic acid 0.01-0.34 mg/g, folic acid 0-0.58 mg/g, vanillic acid 0.05-0.09 mg/g, ferulic acid 0.23-0.76 mg/g, butylidenephthalide 0.06-1.53 mg/g, ligustilide 6.90-24.95 mg/g and butylphthalide 0.36-3.08 mg/g. The contents of commerical Ligustici Rhizoma were as follows: nicotinic acid 0-0.42 mg/g, protocatechuic acid 0.01-0.37 mg/g, phthalic acid 0.09-0.50 mg/g, folic acid 0-0.52 mg/g, p-hydroxybenzoic acid 0-0.03 mg/g, vanillic acid 0.06-0.20 mg/g, caffeic acid 0.01-0.23 mg/g, ferulic acid 0.41-1.10 mg/g, butylphthalide 0.04-0.97 mg/g, senkyunolide A 0.80-3.70 mg/g, ligustilide 6.20-14.06 mg/g and butylidenep-hthalide 3.11-5.74 mg/g.As a result, the presence of senkyunolide A can be used to distinguish Angilicae Radix from Ligustici Rhizoma and the external appearance can be used to predict the quality of the drugs.
