本研究以自營性單細胞藻Isochrysis galbana為實驗藻株(取自東港水產試驗所分離保存的藻株),初期以杯瓶培養,改變光照強度(3000 lux、5000 lux、9000 lux),前培養光照條件(5000 lux��3000 lux、9000 lux��5000 lux、3000 lux��9000 lux),培養溫度(20℃、25℃、30℃) 以找出較佳之培養條件,期望得到較高的生質量及DHA產量。實驗結果顯示,光照5000 lux DHA產量最高達3.93 mg/L,而改變前培養光照條件下,顯示從較差的培養條件轉殖至較好的培養條件,微藻較易適應。最適溫度為25℃,DHA產量達3.21 mg/L。進而擴大到5 L光生化反應器,進行通氣培養。改變光照強度(3000 lux、7000 lux、9000 lux)、通氣量(125 ml/min、250 ml/min、350 ml/min)、循環量(0.5 L/min、1 L/min、3 L/min)來培養,以找出較佳之培養條件,期望得到較高的生質量及DHA產量。實驗結果顯示,光照強度為7000 lux,通氣量為250 ml/min,循環量為1 L/min時DHA產量最高,可達5.68 mg/L。
An autotrophic unicell-algae Isochrysis galbana,obtained from Tong Kung Marine Laboratory,was the strain used in this study Initially,the strain was cultivated in a flask. The objective of this study was to obtain the optimum operating condition for the yield of biomass and DHA via changing the intensity of light(3000 lux、5000 lux、9000 lux),light of preculture(5000 lux��3000 lux、9000 lux��5000 lux、3000 lux��9000 lux),culture temperature(20℃、25℃、30℃) The results indicated that we attained the highest yield of DHA was 3.93 mg/L,which was attained when the intensity of light was 5000 lux,and under changing the preculture condtions, we know that Microalgaes are adapt to the enviorment which transfer worse conditiond to better condition .The fitness temperature is 25℃ and the DHA yield amount to 3.21 mg/L.
