摘 要
1.本研究之目的主在探討中藥方劑作用於實驗動物之消炎、解熱、鎮痛之效果及自發運動之影響,並測定腦內Dopamine(DA),Serotonin (5-HT), Nor-epinephrine(NE)之單胺類(monoamines)含量之變化,用以探討中藥方劑解熱作用的機轉。本實驗使用的中藥方劑有:小柴胡湯( Sheau Chair Hwu Tang ; S.C.H.T : 傷寒論 ) 、十神湯( Shyr Shern Tang ; S.S.T : 太平惠民和劑局方 ) 、九味羌活湯( Zeou Wei Chang Hwo Tang ; Z.W.C.H.T :中國醫學大辭典 )。給藥劑量分別為250mg/kg、500mg/kg、1000 mg/kg,分別以口服給藥法(P.O)投予。口服蒸餾水之組群為控制組 , Aspirin 500mg/kg 為對照組。
實驗結果顯示:在自發運動之影響,在60分十神湯為興奮作用 ,九味羌活湯250mg/kg劑量時為興鐘至180分鐘期間,小柴胡湯1000mg/kg較有顯著抑制作用 ,奮作用,500mg/kg劑量時為抑制作用,1000mg/kg劑量時為興奮作用。 在消炎抗腫脹作用方面十神湯1000mg/kg 與九味羌活湯1000mg/kg 消炎作用呈現近似Aspirin之較佳抗發炎作用。 在解熱作用方面三種方劑在高劑量組1000mg/kg 均有明顯之解熱作用,又以小柴胡湯表現最為顯著。在鎮痛作用方面十神湯與九味羌活湯皆有鎮痛效果呈現以十神湯有優於Aspirin 500mg/kg鎮痛效果 。腦內單胺量之測定項目中 ,小柴胡湯250mg/kg在下視丘DA ( p<0.001 ) 之降低, NE(p<0.05)之降低 , 5-HT (p<0.01 ) 之降低 , 500mg/kg於下視丘DA (p<0.001) 之降低 , 1000mg/kg在下視丘DA ( p<0.001)有意義之降低 ;十神湯1000mg/kg在下視丘5-HT (p<0.01 )降低 ; 由以上實驗結果得知,本研究中所使用的方劑具有良好之消炎、解熱、鎮痛,對自發運動亦無不良影響,各方劑對腦中單胺含量變化之檢測,經由HPLC之測定可以推論出本研究中所使用方劑的解熱作用是透過下視丘單胺神經元的投射作用影響體溫的調節。
Abstract
In this study , we use male Wistar rats, male ICR mice and male New Zealand rabbits as the experiment animals to carry out the following tests: motor activity, anti-swelling effects , antipyretic effects and analgesia effects. We also use high performance liquid chromatography (HPLC) with UV detection to analyze the change volume of monoamines :dopamine(DA), serotonin( 5-HT) and norepinephrine( NE) in brain, which is affected by drugs and compare with physiological response to explore the mechanisms of anti-swelling effects, antipyrexia and analgesia of Sheau Chair Hwu Tang (S.C.H.T), Shyr Shern Tang (S.S.T), Zeou Wei Chang Hwo Tang (Z.W.C.H.T). These three representative formulations administered to test animals orally at doses of 250, 500 and 1000 mg/kg. Control animals were given distilled water, Aspirin served as positive control. Results indicated that (1)S.C.H.T. at dose of 1000 mg/kg during 60~180 minutes and Z.W.C.H.T. at dose of 500 mg/kg significantiy inhibited the of ambulation, (2)S.S.T. at dose of 1000mg/kg and Z.W.C.H.T. at dose of 1000 mg/kg had significant anti-swelling effects, (3)S.C.H.T., S.S.T. and Z.W.C.H.T. at dose of 1000 mg/kg had significant antipyretic effects, (4)S.S.T. and Z.W.C.H.T. the all dose had significant analgesic effects and S.S.T. was better than Aspirin 500mg/kg,(5)S.C.H.T. at dose of 250mg/kg lowered DA(p<0.001), NE(p<0.05) and 5-HT(p<0.01). At 500mg/kg lowered DA(p<0.001), at 1000mg/kg lowered DA (p<0.001). S.S.T. at dose of 1000mg/kg lowered 5-HT (p<0.01) in hippothalamus. From these results , we can find that these three traditional Chinese medicine indeed have good anti-swelling , antipyretic and analgesic effects and correlation between these effects and central neurotransmitters is worthy of our further investigations.
