原子筆墨水主要由染料、樹脂及載體所構成,隨著紙張上墨跡的自然老化,載體成分會揮發逸散,樹脂會聚合,染料成分則發生降解。即使載體成分隨時間逸散,染料成分依然可在墨跡中留存數十年之久,因此染料成分極適合用於原子筆墨水之區分及書寫時間判定之研究。
本研究之目的在評估並尋求最適合原子筆墨跡萃取液之薄層層析法 (TLC)及高效能液相層析法(HPLC)分離系統,並研究峰高比值法(peak ratio methods)應用於書寫時間判定之可行性。本研究選用十隻不同廠牌的藍色、黑色原子筆作為研究對象,為模仿墨跡之自然老化過程,本研究引入人工老化技術(artificial aging technique),墨跡於強烈日光下曝曬不等時間,繼而以甲醇萃取而得不同老化時間之墨跡萃取液樣品。墨跡萃取液樣品先以TLC進行分析,適用之TLC展開劑系統為n-butanol / ethanol/ water = 50: 10: 15 (v/v),藉觀察TLC板上色帶(band)之Rf值及顏色,可發現不同廠牌原子筆墨水間的差異,而新鮮及老化樣品間的變化也可輕易辨別。
本研究使用HPLC伴隨陣列式光電二極體偵檢器(photodiode array detector; PDA detector),檢測不同墨水染料之吸收波長。最適合原子筆墨跡萃取液之HPLC分離系統為THF / acetonitrile / H2O+5mM pic B-7 = 10 / 73 / 17 (v/v)。觀察HPLC層析圖可發現老化樣品會較新鮮樣品產生較多的樣品峰,記錄新鮮樣品及老化樣品中最大吸收波長相同之樣品峰及其峰高值(peak height),計算任兩樣品峰之峰高比值(peak ratio)並繪製峰高比值曲線圖,由圖中可發現新鮮樣品及不同老化時間樣品之曲線變化。藉觀察單一峰高比值曲線,可清楚發現單一峰高比值與不同老化時間之線性趨勢,因此峰高比值法具有極大潛力可應用於原子筆墨水書寫時間之判定。
